The methods involve selecting a bacterial strain having or suspected of having at least one pesticide-encoding plasmid, curing the at least one pesticide-encoding plasmid from the bacterial strain and performing a subtractive hybridization to obtain plasmid mRNA encoding the pesticidal polypeptide.
