  • Publication number: 20200354701
Abstract: The embodiments disclosed herein utilize fluorescence polarization based preliminary screen to identify a putative set of Cas inhibitors from an initial set of candidate inhibitors. The primary screening assay is followed by secondary screening assay to validate the putative set of inhibitors selected by the preliminary screen. In some embodiments, the present disclosure includes compositions and methods are provided for the inhibition of the function of RNA guided endonucleases, including the identification and use of such inhibitors.

  • Publication number: 20200239879
Abstract: Compositions and methods for conditionally regulating activities of CRISPR-Cas systems. In some embodiments, the methods comprise providing an inactive guide RNA comprising a regulatory domain bound by a lock nucleic acid different from the guide RNA; and displacing the lock nuclei acid from the regulatory domain by a trigger nucleic acid, thereby activating the guide RNA, wherein the activated guide RNA forms a complex with a Cas enzyme.

  • Publication number: 20200032322
Abstract: A method for identifying a nucleic acid template that include (a) providing a plurality of primer-template hybrids, wherein a first hybrid of the plurality includes a first template hybridized to a first primer, and wherein a second hybrid of the plurality includes a second template hybridized to a second primer, the second primer having a ternary complex inhibitor moiety at the 3? end; (b) delivering polymerases and nucleotides to the plurality, whereby the first hybrid binds a polymerase and nucleotide to form a stabilized ternary complex and whereby the second hybrid does not bind a polymerase and nucleotide to form a stabilized ternary complex; and (c) detecting the stabilized ternary complex to identify the first template.

  • Patent number: 10467035
Abstract: Techniques are described for importing and using virtual machine images in configured manners, such as by a virtual machine image importation service on behalf of clients. An image may be retrieved based on a client-provided location, and various characteristics of the image may be identified via application of multiple heuristic or other assessment tests to various aspects of the retrieved image such as a master boot record, filesystem, or directory structure of the image to determine a testing vector that is compared to identification vectors associated with known operating systems, in order to automatically determine a specific operating system installed on the image to be imported. Modifications may be made to drivers, configuration and system files of the retrieved image based on the identified operating system and other image characteristics.

Date of Patent: November 5, 2019
    Assignee: Amazon Technologies, Inc.
Inventors: Ekanth Sethuramalingam, Amita Ekbote, Hari Subramanian, Nagaraju Shiramshetti, Sudharsan Balakrishnan Sripadham, Raviprasad Venkatesha Murthy Mummidi, Sophia Yeemei Tsang
