    Abstract: A bioreactor has a cover, a container body, a sealing portion, a driving portion and a liquid guide portion. A ventilation structure and a bacteria-retaining sealing breathable structure provided in the cover, a cleaning liquid inlet portion penetrates the bottom of the driving portion, and a liquid inlet portion and a liquid outlet portion of the liquid guide portion are arranged penetrating a sidewall of the container body. The driving portion drives, by means of elastic deformation, a liquid to be replaced or a cleaning liquid to move up and down, such that the liquid to be replaced can enter the container body only through a gas-liquid channel, so as to subsequently prevent damage to hepatic cells due to a carrier bearing a shearing force, and enable the liquid to be replaced to gain full contact with the carrier. Further provided is a bioreaction system.

    Abstract: The invention provides a method for constructing a hepatic progenitor cell-like cell bank, including successively performing following processes to human primary hepatocyte cultures from different donor sources: transformation-culture, cryopreservation treatment, proliferation-culture, a first subculture treatment, virus infection, a second subculture treatment, continuous selection-culture and continuous subculture. In the method for constructing a heterogenous immortal hepatic progenitor cell-like cell bank of the present invention, the human primary hepatocyte culture of each of the donor sources is transformation-cultured before the proliferation-culture, which is beneficial in endowing the human primary hepatocyte cultures with good proliferation performance. Once combined with subsequent controlling of culture parameter, the immortal hepatic progenitor cell-like cell lines obtained from different donor sources may have good in vitro proliferation ability.

    Abstract: A method of constructing a hepatic precursor-like cell line is provided. The deposited number of the hepatic precursor-like cell line is CCTCC NO: C2019120. The construction method includes using primary hepatocytes as seed cells to establish the hepatic precursor-like cell line, which combines with controlling the conditions of passage culture, subculture, virus infection, proliferation culture and confluence culture. Moreover, a hepatocyte-specific transcription factor, FOXA3, is expressed in the immortalized cell line to make the hepatic precursor-like cell line differentiate easily. An application of the hepatic precursor-like cell line in the field of the bioartificial liver is also provided.
