    Abstract: An optical fiber transducer usable in environments of extreme operating temperature features a stationary support, a movable body displaceable back and forth relative thereto, and an optical fiber connected between the support and the movable body. The fiber has a Fiber Bragg Grating in an intermediate region thereof between the support and movable body. To accommodate varying coefficients of thermal expansion (CTEs) among these components, one or more tubes close circumferentially around the fiber. Each tube has a CTE that is greater than that of the fiber, and less than that of the constituent material of the support and movable body. The fiber is bonded to an interior of the tube(s), while an exterior of the tube(s) is bonded to the support and movable body.

    Abstract: The present disclosure relates to methods and systems for realization of a reference pressure as well as calibration of devices under test. The techniques leverage the measurement of buoyancy artifacts under vacuum and pressure conditions, and the use of gas law equations and related variables to obtain low uncertainty reference values for pressure among others. The techniques can include measuring an absolute mass difference of buoyancy artifacts under vacuum; measuring effective masses of the buoyancy artifacts under a gas pressure condition, and determining an effective mass difference between the buoyancy artifacts; and determining a low-uncertainty pressure based on the absolute mass difference, effective mass difference, Boltzmann constant, volume difference, molecular weight of the gas at pressure, and temperature of the measurements.

    Abstract: Provided are methods of producing target capture nucleic acids. The methods comprise bidirectionally amplifying a circular nucleic acid template by rolling circle amplification (RCA) using first and second primers, where the circular nucleic acid template comprises a target nucleotide sequence and a restriction site. The bidirectional amplification produces a double-stranded concatemer comprising a first strand comprising a plurality of linked units, each unit comprising the target nucleotide sequence and the restriction site, and a second strand which is the reverse complement of the first strand. The methods further comprise digesting the double-stranded concatemer using a restriction endonuclease that cleaves the restriction site to produce a plurality of restriction fragments. Also provided are target capture nucleic acids produced according to such methods. Methods of capturing target nucleic acids using target capture nucleic acids produced according to such methods are also provided.

In some embodiments, the methods include producing a concatemer by rolling circle amplification (RCA) of a circular nucleic acid template, where the circular nucleic acid template includes a barcode and a stem-loop forming region, and where the concatemer includes a plurality of linked units, each unit including the barcode and a stem-loop structure formed from the stem-loop forming region. Such methods further include disposing the concatemer on a solid support to produce a barcoded solid support including a plurality of the stem-loop structures extending from the surface of the solid support. The methods may further include treating the stem-loop structures with an agent that produces stem structures having ends compatible with target nucleic acids, and attaching the target nucleic acids to the stem structures. Barcoded solid supports and methods of using the barcoded solid supports are also provided.
