    Abstract: Pharmacologically active, radioactive, and photoactive vinblastine analogs are provided which can be used to bind covalently to cellular polypeptides which have high affinity for Vinca alkaloids. The compounds are N-(p-azido[3,4-.sup.3 H]benzoyl)-N'beta-aminoethylvindesine and N-p-azido-[3-.sup.125 I]-salicyl-N'-beta-aminoethylvindesine. The compounds can be used to identify cellular Vinca alkaloid receptors which may be involved in antineoplastic, cytotoxic and drug resistant mechanisms of actions. In addition to specific interaction with tubulin, these compounds specifically bind to a 150-180 kDa surface membrane glycoprotein which is overexpressed in multidrug resistant cells.

    Abstract: A technique for identifying an unknown microorganism, in which technique a specimen of the unknown microorganism is prepared to which a radioactive emissive agent has been added that is actively incorporated into the products of metabolism of the microorganism to produce a mix of radioactive peptide or protein emissive products in a manner that depends on the metabolic mechanism of the microorganism. The peptide or protein emissive products in the mix resulting from the preparation of the specimen are then separated. The separated emissive products are detected by sensing the emission from the products to provide an electrical signal whose wave pattern is indicative of the identity of the microorganism. This wave pattern is compared in a computer with patterns stored therein representing a collection of known microorganisms.

    Abstract: A method for the early diagnosis of Bordetella diseases based on the detection of Bordetella adenylate cyclase and the ability of this enzyme to catalyze the formation of cyclic adenosine monophosphate (cAMP). A sample secretion is taken from a suspected victim of a Bordetella disease (the most common of which in humans is whooping cough) and incubated under conditions to promote the formation of cAMP, which may then be measured as evidence of the presence of the disease. A sample of nasopharyngeal secretions is maintained in a nutrient medium pending assay. An adenylate cyclase assay is made by incubating the sample with adenosine triphosphate (ATP) and the calcium binding protein calmodulin to generate cAMP. The cAMP is measured by any of several known available methods.

    Abstract: A fast growing, continuous, functional rat thyroid cell strain, FRTL-5, which maintains functional characteristics of iodide uptake and thyroglobulin synthesis over prolonged periods of culture is cloned from FRTL cells obtained from primary cultures of Fischer rat thyroid glands. The FRTL-5 cells are cultured in a medium containing approximately 5 percent calf serum supplemented with a mixture of hormones, at least one of which is thyrotropin.The FRTL-5 cells are employed in a series of assays which measure thyroid stimulatory or inhibitory factors. The FRTL-5 system of assay specifically measures thymidine incorporation, cAMP elevation and iodide uptake and permits the evaluation of patient sera, particularly those afflicted with Graves' disease and other autoimmune thyroid diseases, thereby providing a means for determining appropriate methods of treatment.

The corresponding polyurethane(urea) compositions contain up to 95% by weight of lignite and/or peat in the filler-containing polyurethane(urea) composition and are already swollen from production. The water absorbability (WAF) value thereof when suspended in water is from 33 to 97% by weight of water.

    Abstract: An identification technique for micro-organisms in which a dilute solution of a culture medium containing an unknown micro-organism has added thereto an emissive agent such as a radioactive amino acid to produce a mix of emissive products that depends on the metabolic mechanism of the micro-organism. After a predetermined incubation period, the reaction is arrested and the solution layered onto a gel plate where it is subjected to electrophoresis. The plate is then autoradiographed by exposing the gel to a sensitive photographic film for a period sufficient to produce thereon a characteristic band pattern functioning as an identifier for the micro-organism. Identification may be effected by comparing the identifier for the unknown with a collection of identifiers for known micro-organisms to find a match with one of these known identifiers.

    Abstract: Method, apparatus and automated system for determining the effects of a chemical agent upon a bacterial sample. Aliquots of a bacterial suspension in a culture medium, the chemical agent, and tritiated thymidine are deposited in polyethylene tubes having first and second compartments separated by a fixed millipore filter. A precipitation agent, such as trichloroacetic acid, is deposited into the polyethylene tubes to precipitate the bacteria. The bacteria is separated from the liquid by operation of a vacuum system which draws the liquid through the filter, thereby filtering out the bacteria. The filtered bacteria is subsequently washed with a washing solution. In one embodiment, a scintillation fluid is added to the bacteria, the bacteria then being conveyed to a scintillation counter which produces an output signal corresponding to the amount of radiation emanating from the bacteria.

    Abstract: Method and compositions for infectious disease diagnosis and epidemiology involving labeled nucleotide probes complementary to nucleic acid coding for a characteristic pathogen product. Clinical isolates are cultivated, expanding the number of microorganisms, the resulting colonies lysed, the genome normally denatured and then fixed. Alternatively, clinical samples (stool, sputum, pus, etc.) are spotted onto an inert support. The sample is treated in such a way that the DNA is liberated from microbes present in the sample and complexed onto the support. The DNA is normally denatured and fixed in this process. Subsequently, a labelled polynucleotide probe specific for a DNA sequence characteristic of a pathogenic product suspected of being present in the clinical sample is contacted with the fixed genomic single stranded nucleic acid under hybridizing conditions. Hybridization of probes to the single stranded nucleic acid is diagnostic of the presence of the pathogen.

    Abstract: The present invention provides a test sample container for measuring the biological activity of cells, bacteria, or micro-organisms in a test sample by radiometric techniques. The container is suited for use with automated laboratory equipment and provides greatly improved measurement efficiency over prior designs. In one embodiment, the test sample container includes a serum vial adapted to contain a quantity of a test sample and a radionuclide-labeled nutrient media. A micro-tube, which contains a mixture of a gas absorber and a scintillation compound, is positioned above the surface of the sample/media mix by a split-skirt stopper that also serves to seal the serum vial. The gas metabolized or otherwise evolved by the micro-organisms in the test sample is absorbed by the absorber in the micro-tube causing the scintillation compound to emit light flashes at a rate proportional to the quantity of radioactive gas absorbed.
