Patent number: 6485915Abstract: The present invention concerns a method for the detection of nucleic acids on an analytical element which contains a sample application zone and a detection zone, the analytical element enabling liquid transport from the sample application zone to the detection zone, wherein a sample is applied to the sample application zone of the analytical element and the nucleic acids contained in the sample are denatured and can be detected qualitatively in the detection zone by hybridization with a detection probe and also quantitatively, preferably by means of marker groups. In addition new analytical elements and reagent kits for the detection of nucleic acids are provided.Type: GrantFiled: November 17, 2000Date of Patent: November 26, 2002Assignee: Roche Diagnostics GmbHInventors: Volker Keller, Andreas Rauscher, Joachim Steinbiss, Reiner Schlipfenbacher, Jurgen Klepp

Patent number: 6207000Abstract: A process for the production of analytical devices is provided in accordance with the present invention. Analytical devices include analytical test elements with a capillary-active zone for examining liquid samples. In the process a carrier layer is prepared, a spacer layer is laminated onto the carrier layer, a contour is punched, cut or stamped through the spacer layer laminated onto the carrier layer which determines the shape of the capillary-active zone, those parts of the spacer layer which are not required to form the capillary-active zone are removed from the carrier layer and a cover layer is applied to the spacer layer to result in a capillary-active zone. The process according to the invention is preferably suitable for manufacturing analytical devices from tape material.Type: GrantFiled: April 1, 1999Date of Patent: March 27, 2001Assignee: Roche Diagnostics GmbHInventors: Wolfgang Schwobel, Bernd Hein

Patent number: 5989888Abstract: A purified enzyme mixture useful for isolating cells or cell clusters from tissue is disclosed. The mixture includes at least two collagenase enzymes, at least two other proteases, and additional non-protease components. The mixture is purified by removing at least some of the non-protease components. The purified mixture may then be used to isolate cells or cell clusters from tissue. Also disclosed are the essential components of the purified enzyme mixture as well as preferred ranges and ratios of these essential components for isolating cells or cell clusters from tissue. Finally, a dissociation system is disclosed that can be used with the purified enzyme mixture to dissociate tissue and recover cells or cell clusters.Type: GrantFiled: March 13, 1998Date of Patent: November 23, 1999Assignee: Roche Diagnostics CorporationInventors: Francis E. Dwulet, Bernice B. Ellis, John F. Gill, Linda B. Jacobsen, Marilyn E. Smith, David G. Waters

Patent number: 5846702Abstract: A new group of Os(II) and Os(III) compounds useful as redox mediators in electrochemical biosensors. These compounds have 1) low oxidation potential, 2) fast reaction kinetics between the electroactive center of an enzyme and the compound, 3) slow oxidation of osmium by oxygen, and 4) excellent solubility in aqueous medium. These mediators are particularly useful as a component of a reagent used in an electrochemical biosensor, wherein the biosensor is useful for measuring analytes from a biological fluid, such as blood.Type: GrantFiled: December 20, 1996Date of Patent: December 8, 1998Assignee: Boehringer Mannheim CorporationInventors: Zhi David Deng, Gerald F. Patent number: 5830741Abstract: An enzyme composition for dissociating tissues is disclosed. The composition comprises purified collagenase I and collagenase II from Clostridium histolyticum in a mass ratio of about 0.3 to 0.6 and a neutral protease such that the ratio of total FITC casein units of activity to Wunsch units of activity in the composition is about 250:1 to about 600:1. The preferred neutral protease is thermolysin.Type: GrantFiled: December 6, 1996Date of Patent: November 3, 1998Assignee: Boehringer Mannheim CorporationInventors: Francis E. Dwulet, Marilyn E. Smith

Patent number: 5762770Abstract: An electrochemical biosensor test strip that has a minimum volume blood sample requirement of about 9 microliters. The test strip has working and counter electrodes that are substantially the same size and made of the same electrically conducting material placed on a first insulating substrate. Overlaying the electrodes is a second insulating substrate, which includes a cutout portion that forms a reagent well. The cutout portion exposes a smaller area of the counter electrode than the working electrode. A reagent for analysis of an analyte substantially covers the exposed areas of working and counter electrodes in the reagent well. Overlaying the reagent well and affixed to the second insulating substrate is a spreading mesh that is impregnated with a surfactant. The small cutout portion of 4 millimeters by 4.2 millimeters, small mesh of 6 millimeters by 5.8 millimeters, and small amount of reagent, 4 microliters before drying, allow the test strip to analyze a whole blood sample of about 9 microliters.Type: GrantFiled: June 30, 1995Date of Patent: June 9, 1998Assignee: Boehringer Mannheim CorporationInventors: G.
