    Abstract: An optical probe enables the study of enzyme activity by absorbance spectroscopy or by sensitive fluorescence methods. In particular, the probe provides the ability to monitor the activity of cytochrome P-450.sub.scc enzyme, the rate limiting enzyme for steroid biosynthesis. Located on the inner mitochondrial membrane, P-450.sub.scc catalyzes the conversion of cholesterol to pregnenolone and isocapraldehyde by sequential oxidations of the cholesterol side chain. The fluorogenic probe includes a cholesterol-like steroid linked to a chromophore through a linking group. The chromophore is selected to have little optical response when linked to the steroid substrate and an enhanced optical response when cleaved from the substrate and linking group. Thus, a fluorescent anion that can be optically detected is generated by the side-chain cleavage reaction during steroidogenesis.

    Abstract: A feedthrough bushing for a high voltage diode provides for using compression sealing for all sealing surfaces. A diode assembly includes a central conductor extending through the bushing and a grading ring assembly circumferentially surrounding and coaxial with the central conductor. A flexible conductive plate extends between and compressively seals against the central conductor and the grading ring assembly, wherein the flexibility of the plate allows inner and outer portions of the plate to axially translate for compression sealing against the central conductor and the grading ring assembly, respectively. The inner portion of the plate is bolted to the central conductor for affecting sealing. A compression beam is also bolted to the central conductor and engages the outer portion of the plate to urge the outer portion toward the grading ring assembly to obtain compression sealing therebetween.

    Abstract: A high resolution event interval timing system measures short time intervals such as occur in high energy physics or laser ranging. Timing is provided from a clock (38) pulse train (37) and analog circuitry (44) for generating a triangular wave (46) synchronously with the pulse train (37). The triangular wave (46) has an amplitude and slope functionally related to the time elapsed during each clock pulse in the train. A converter (18, 32) forms a first digital value of the amplitude and slope of the triangle wave at the start of the event interval and a second digital value of the amplitude and slope of the triangle wave at the end of the event interval. A counter (26) counts the clock pulse train (37) during the interval to form a gross event interval time. A computer (52) then combines the gross event interval time and the first and second digital values to output a high resolution value for the event interval.

    Abstract: A climbing apparatus is provided for climbing ferromagnetic surfaces, such as storage tanks and steel frame structures. A magnet assembly is rotatably mounted in a frame assembly. The frame assembly provides a pair of cam surfaces having different dimensions so that, when the frame is rotated, the cam surfaces contact the ferromagnetic surface to separate the magnet assembly from the surface. The different cam dimensions enable one side of the magnet at a time to be detached from the surface to reduce the effort needed to disengage the climbing apparatus. The cam surface also provides for smoothly attaching the apparatus. A hardened dowel pin is also attached to the frame and the pointed end of the dowel engages the surface when the magnet is attached to the surface to prevent downward sliding movement of the assembly under the weight of the user.

    Abstract: Fading channel effects on a transmitted communication signal are simulated with both frequency and time variations using a channel scattering function to affect the transmitted signal. A conventional channel scattering function is converted to a series of channel realizations by multiplying the square root of the channel scattering function by a complex number of which the real and imaginary parts are each independent variables. The two-dimensional inverse-FFT of this complex-valued channel realization yields a matrix of channel coefficients that provide a complete frequency-time description of the channel. The transmitted radio signal is segmented to provide a series of transmitted signal and each segment is subject to FFT to generate a series of signal coefficient matrices. The channel coefficient matrices and signal coefficient matrices are then multiplied and subjected to inverse-FFT to output a signal representing the received affected radio signal.

    Abstract: A G.sub.1 phase kinase inhibitor is applied in a low concentration to a population of normal and transformed mammalian cells. The concentration of G.sub.1 phase kinase inhibitor is selected to reversibly arrest normal mammalian cells in the G.sub.1 cell cycle without arresting growth of transformed cells. The transformed cells may then be selectively identified and/or cloned for research or diagnostic purposes. The transformed cells may also be selectively killed by therapeutic agents that do not affect normal cells in the G.sub.1 phase, suggesting that such G.sub.1 phase kinase inhibitors may form an effective adjuvant for use with chemotherapeutic agents in cancer therapy for optimizing the killing dose of chemotherapeutic agents while minimizing undesirable side effects on normal cells.

    Abstract: A flat panel display incorporates a plurality of micro-pump light valves (MLV's) to form pixels for recreating an image. Each MLV consists of a dielectric drop sandwiched between substrates, at least one of which is transparent, a holding electrode for maintaining the drop outside a viewing area, and a switching electrode from accelerating the drop from a location within the holding electrode to a location within the viewing area. The sustrates may further define non-wetting surface areas to create potential energy barriers to assist in controlling movement of the drop. The forces acting on the drop are quadratic in nature to provide a nonlinear response for increased image contrast. A crossed electrode structure can be used to activate the pixels whereby a large flat panel display is formed without active driver components at each pixel.

    Abstract: A cryogenic refrigerator cools a heat source by cyclically concentrating and diluting the amount of .sup.3 He in a single phase .sup.3 He-.sup.4 He solution. The .sup.3 He in superfluid .sup.4 He acts in a manner of an ideal gas in a vacuum. Thus, refrigeration is obtained using any conventional thermal cycle, but preferably a Stirling or Carnot cycle. A single phase solution of liquid .sup.3 He at an initial concentration in superfluid .sup.4 He is contained in a first variable volume connected to a second variable volume through a superleak device that enables free passage of .sup.4 He while restricting passage of .sup.3 He. The .sup.3 He is compressed (concentrated) and expanded (diluted) in a phased manner to carry out the selected thermal cycle to remove heat from the heat load for cooling below 1 K.

    Abstract: The resonance frequency spectrum of a magnetostrictive sample is remotely determined by exciting the magnetostrictive property with an oscillating magnetic field. The permeability of a magnetostrictive material and concomitant coupling with a detection coil varies with the strain in the material whereby resonance responses of the sample can be readily detected. A suitable sample may be a magnetostrictive material or some other material having at least one side coated with a magnetostrictive material. When the sample is a suitable shape, i.e., a cube, rectangular parallelepiped, solid sphere or spherical shell, the elastic moduli or the material can be analytically determined from the measured resonance frequency spectrum. No mechanical transducers are required and the sample excitation is obtained without contact with the sample, leading to highly reproducible results and a measurement capability over a wide temperature range, e.g.

    Abstract: A microwiggler assembly produces large magnetic fields for oscillating ched particle beams, particularly electron beams for free electron laser (FEL) application. A tube of electrically conductive material is formed with radial slots axially spaced at the period of the electron beam. The slots have alternate 180.degree. relationships and are formed to a maximum depth of 0.6 to 0.7 times the tube circumference. An optimum slot depth is selected to eliminate magnetic quadrupole fields within the microwiggler as determined from a conventional pulsed wire technique. Suitable slot configurations include single slits, double slits, triple slits, and elliptical slots. An axial electron beam direction is maintained by experimentally placing end slits adjacent entrance and exit portions of the assembly, where the end slit depth is determined by use of the pulsed wire technique outside the tube.

In a particular application, the color values in an image are compressed for storage in a discrete lookup table (LUT) where an 8-bit data signal is enabled to form a display of 24-bit color values. The LUT is formed in a sampling and averaging process from the image color values with no requirement to define discrete Voronoi regions for color compression. Image color values are assigned 8-bit pointers to their closest LUT value whereby data processing requires only the 8-bit pointer value to provide 24-bit color values from the LUT.

    Abstract: An RF transport system delivers a phase-stable RF signal to a load, such as an RF cavity of a charged particle accelerator. A circuit generates a calibration signal at an odd multiple frequency of the RF signal where the calibration signal is superimposed with the RF signal on a common cable that connects the RF signal with the load. Signal isolating diplexers are located at both the RF signal source end and load end of the common cable to enable the calibration to be inserted and extracted from the cable signals without any affect on the RF signal. Any phase shift in the calibration signal during traverse of the common cable is then functionally related to the phase shift in the RF signal. The calibration phase shift is used to control a phase shifter for the RF signal to maintain a stable RF signal at the load.

    Abstract: An integrated fluorescence analysis system enables a component part of a sample to be virtually sorted within a sample volume after a spectrum of the component part has been identified from a fluorescence spectrum of the entire sample in a flow cytometer. Birefringent optics enables the entire spectrum to be resolved into a set of numbers representing the intensity of spectral components of the spectrum. One or more spectral components are selected to program a scanning laser microscope, preferably a confocal microscope, whereby the spectrum from individual pixels or voxels in the sample can be compared. Individual pixels or voxels containing the selected spectral components are identified and an image may be formed to show the morphology of the sample with respect to only those components having the selected spectral components. There is no need for any physical sorting of the sample components to obtain the morphological information.

    Abstract: Two betatrons are provided in tandem for alternately accelerating an electron beam to avoid the single flux swing limitation of conventional betatrons and to accelerate the electron beam to high energies. The electron beam is accelerated in a first betatron during a period of increasing magnetic flux. The eletron beam is extracted from the first betatron as a peak magnetic flux is reached and then injected into a second betatron at a time of minimum magnetic flux in the second betatron. The cycle may be repeated until the desired electron beam energy is obtained. In one embodiment, the second betatron is axially offset from the first betatron to provide for electron beam injection directly at the axial location of the beam orbit in the second betatron.

A manipulation chamber provides a plurality of inlet and outlet ports for the particles and for fluids used to control or to contact the particles. A central manipulation area is optically accessible by the laser and includes first enlarged volumes for containing a selected number of particles for experimentation. A number of first enlarged volumes are connected by flow channels through second enlarged volumes. The second enlarged volumes act as bubble valves for controlling the interconnections between the first enlarged volumes. Electrode surfaces may be applied above the first enlarged volumes to enable experimentation using the application of electric fields within the first enlarged volumes.
