Patent number: 5223407Abstract: A recombinant DNA construct, useful for obtaining excretion from E. coli of a heterologous protein, comprises a coding region in which DNA coding for the heterologous protein in coupled in reading frame with DNA coding for the OmpA signal peptide, to enable secretion of the protein into the periplasm; and a control region linked operably with the coding region to enable expression thereof in an E. coli host, wherein the control region comprises the tac promoter, the lac operator and a consensus ribosome binding site. With this construct, the excretion of heterologous proteins from E. coli can be achieved, consistently and for a wide variety of such proteins, without assistance from carrier proteins or membrane-lytic agent and by using "healthy" hosts rather than mutant strains having defective outer membranes.Type: GrantFiled: August 18, 1989Date of Patent: June 29, 1993Assignee: Allelix Inc.Inventors: W. Patent number: 4885249Abstract: Transformants of Aspergillus niger and related Aspergilli, containing foreign DNA conferring modified properties of expression thereon, are prepared by use of a DNA vector which contains a selectable marker which is capable of incorporation into the DNA of the host A. niger cells, but which is not to be found in the A. niger cells prior to this transformation. The vector also contains other foreign DNA to be incorporated into the A. niger, for modified expression. The process suitably uses mutants of A. niger as hosts, the mutants lacking the selectable marker as compared with wild type A. niger, and conducts the transformation on spheroplasts of A. niger.Type: GrantFiled: December 5, 1984Date of Patent: December 5, 1989Assignee: Allelix, Inc.Inventors: Franics P. Buxton, David I. Gwynne, Roger W. Davies

Patent number: 4847203Abstract: A fermentation vessel comprises an outer shell, and an annular draft tube located within the shell and spaced therefrom to promote continuous circulation of fluids through said draft tube within said shell. The draft tube has an internal cavity located within its walls. The cavity is connected to a fluid supply conduit that extends externally of the shell. A plurality of discrete nozzles are disposed about the circumference of the tube and communicate with the cavity to permit egress of fluids from the cavity to the interior of the shell to promote circulation.Type: GrantFiled: August 31, 1987Date of Patent: July 11, 1989Assignee: Allelix, Inc.Inventor: Nigel J. Smart

Patent number: 4831133Abstract: Alkaloids present in Catharanthus roseus tissue are selectively concentrated by aqueous extraction of ground tissue followed by extraction with organic solvent e.g. ethyl acetate. The resultant concentrate is rich in catharanthine, vindoline and 3',4'-anhydrovinblastine (AVLB). Yield of AVLB is enhanced by addition to the aqueous extraction medium of acid, salt, or hydrogen peroxide and can be further enhanced through the addition of sodium borohydride.Type: GrantFiled: May 1, 1987Date of Patent: May 16, 1989Assignee: Allelix, Inc.Inventors: Anne E. Goodbody, Colin D. Watson, Masanaru Misawa

Patent number: 4791060Abstract: The invention relates to a device and method for performing qualitative enzyme immunoassays. The device comprises at least one tube having antibody, antigen or hapten attached to an internal surface thereof. A first syringe is connectable to a first end of the tube or tubes and supplies a flow of test liquid therethrough. A second syringe having a two piston arrangement is connected to the tube so as to allow a first wash solution followed by an enzyme conjugate solution to be flowed therethrough. Finally a third syringe, also having a two piston configuration, is connected to the tube so as to allow a second wash solution followed by an enzyme substrate and metabolite indicator solution to be flowed therethrough.Type: GrantFiled: November 21, 1986Date of Patent: December 13, 1988Assignee: Allelix Inc.Inventor: Howard M. Chandler

Patent number: 4729875Abstract: A device for performing a plurality of immunochemical assays simultaneously comprises a waste reservoir about which are positioned at least two assay tubes connected in series by conduits formed in the base and top of the reservoir structure. A compound syringe is insertable into an opening formed through the top of the waste reservoir, the syringe being held in place by a receiving structure formed in the base of the reservoir and connected to the entrance end of the series of assay tubes. The various compartments of the compound syringe are emptied in sequence to cause the flow of sample and assay reagents to flow through the assay tubes. The device is structured to allow for the reflow of sample through the tubes if desired.Type: GrantFiled: June 26, 1986Date of Patent: March 8, 1988Assignee: Allelix Inc.Inventor: Howard M. Chandler
