    Abstract: The invention provides a radiotherapeutical or radiosurgical system comprising at least two high-intensity radiation sources configured to rotate around a common rotation axis and a ring-shaped imaging device. A three-source configuration is considered as the most cost-effective and will be used as an example for illustration. The configuration of these high-intensity radiation sources and use of a unique compact MLC for each of the radiation sources make it possible for the system to rapidly deliver high-conformal non-coplanar stereotactic radiation treatment in one gantry rotation without any couch rotation. Therefore the invention can deliver high precision and high-conformal non-coplanar stereotactic radiation treatment to any part of the body in an extremely short time (0.1-20 seconds), which may exhibit numerous advantages over the prior art, such as reduction of radiation damage to the circulating immune cells in blood and mitigation of patient motion-induced problems, among others.

    Abstract: A method for increasing the proportion of a central memory T cell (Tcm) subset in a CAR-T cell product includes adding an artificial antigenic epitope in a CAR, and activating a CAR-T by the artificial antigenic epitope, wherein the artificial antigenic epitope does not exist in other domains or segments of the CAR. A method for preparing a CAR-T cell includes: introducing the CAR into a T cell; and culturing the CAR-introduced T cell, and performing specific activation in the culture process. The preparation method can not only increase the proportion of CAR-positive T cells in the product, but also can achieve specific in vitro amplification of the Tcm subset in the CAR-positive T cells, and significantly increases the proportion of Tcm in a final product. Clinical trials show that the CAR-T cells have significantly improved amplification capacity in vivo, and improved clinical safety and effectiveness.

    Abstract: The present disclosure provides a fluorescently-traceable amino acid derivative and a preparation method and use thereof, and belongs to the technical field of biomedicine. In the present disclosure, an amino acid skeleton of the amino acid derivative is modified mainly by a fluorescently-traceable functional group. 4-chloro-7-nitro-2,1,3-benzoxadiazole (NBD-Cl) has low polarity and strong fluorescence, and can be used to modify an N-terminal of the amino acid skeleton to conduct subcellular imaging. Moreover, the NBD-Cl has a relatively small volume, lacks reaction orthogonality, and causes little interference with biochemical reactions of the organism itself. The results of examples show that the fluorescently-traceable amino acid derivative has a desirable biological activity and can be fluorescently traced in vivo and in vitro.
