Abstract: Disclosed are methods of treating a subject following a small-volume ischemic stroke suffered by the subject and methods of treating a subject with a stroke-induced motor deficit. Disclosed also is a composition for treating small-volume ischemic stroke. In one aspect, the method of treating a subject following a small-volume ischemic stroke comprises administering, to a brain region surrounding a small-volume ischemic core of the subject, a therapeutically effective amount of cells, wherein the cells are descended from mesenchymal stem cells transiently-transfected by a polynucleotide encoding a Notch intracellular domain.
Abstract: A method of preparing a syringe in connection with a therapeutic treatment is disclosed. The method can include removing a plunger of the syringe from a barrel of the syringe, aligning the barrel in a horizontal orientation, filling a lumen of the barrel with a viscous material through an opening at a proximal end of the barrel, and inserting a plunger tip into the lumen to seal the lumen. The method can also include attaching an implantation device to a hub coupled to the barrel at a distal end of the barrel. The method can also include depressing the plunger until the cell suspension fills the implantation device and a droplet of the cell suspension is expelled from a distal tip of the implantation device.
Abstract: There is provided a method of inducing differentiation of bone marrow stromal cells to neural cells or skeletal muscle cells by introduction of a Notch gene. Specifically, the invention provides a method of inducing differentiation of bone marrow stromal cells to neural cells or skeletal muscle cells in vitro, which method comprises introducing a Notch gene and/or a Notch signaling related gene into the cells, wherein the finally obtained differentiated cells are the result of cell division of the bone marrow stromal cells into which the Notch gene and/or Notch signaling related gene have been introduced. The invention also provides a method of inducing further differentiation of the differentiation-induced neural cells to dopaminergic neurons or acetylcholinergic neurons. The invention yet further provides a treatment method for neurodegenerative and skeletal muscle degenerative diseases which employs neural precursor cells, neural cells or skeletal muscle cells produced by the method of the invention.
