The differentiation-inducing culture medium additive of the present invention does not require ascorbic acid 2-phosphate and ITS, which are normally essential for bone differentiation. This method enables to distinguish an undifferentiated mesenchymal stem cell from other connective tissue cell such as fibroblasts, osteoblasts, chondrocytes and adipose cells with good accuracy. A mesenchymal stem cell given by this method or a composition comprising the mesenchymal stem sell can be used as a therapeutic for use in the regenerative medicine.
