  • Publication number: 20070141368
Abstract: A gas turbine engine nozzle segment and process for producing such a nozzle segment to exhibit improved durability and aerodynamic performance. The process produces a nozzle segment having at least one vane between and interconnecting a pair of platforms. The nozzle segment is cast from a gamma prime-strengthened nickel-base superalloy, on whose surface is thermal sprayed an environmental coating formed of a MCrAlX-type coating material. The surface of the environmental coating is then worked to cause the coating to have a surface finish of less than 2.0 micrometers Ra. Cooling holes are then drilled in the nozzle segment, after which an oxidation-resistant coating is applied on the smoothed surface of the nozzle segment so as to maintain an outermost surface on the nozzle segment having surface finish of less than 2.0 micrometers Ra.

  • Patent number: 7186989
Abstract: The present invention provides core technologies necessary for a portable, low cost, thermal-regulating LED-based handheld fluorometer. The regulated fluorometer is based on a low thermal mass infrared heater, and an orthogonal geometry LED based filter fluorometer. Power is supplied through an external power supply and data is collected in real-time through standard serial interfaces of personal computers or personal digital assistants. Thermal regulation is automatically maintained using temperature sensor feedback control. Optical excitation relies on LED light source(s) and optical detection is through an adjustable integrating photodetector. Such a handheld system can allow applications requiring temperature sensitive photometric measurements for real time analyte detection to be performed in the field.

  • Patent number: 7179644
Abstract: Methods are provided for producing an expression vector. In the subject methods, donor and acceptor vectors are combined in the presence of a recombinase to produce an expression vector that includes a first and second recombinase recognition site oriented in the same direction, wherein the first and second recombination sites are able to recombine with each other. In the subject methods, one of the donor and acceptor vectors includes a single recombinase recognition site while the other includes two recombinase recognition sites. Also provided are compositions for use in practicing the subject methods, including the donor and acceptor vectors themselves, as well as systems and kits that include the same. The subject invention finds use in a variety of different applications, including the transfer or cloning of a nucleic acid of interest from a first vector into one or more expression vectors, etc.

  • Publication number: 20060093849
Abstract: A method for applying a chromium containing coating to an underlying metal substrate where the metal substrate has an overlaying platinum-containing layer, as well as a corrosion resistant coated article thereof. A chromium-containing layer is deposited on the platinum-containing layer with an aluminide diffusion layer being deposited on the chromium-containing layer, the aluminide diffusion layer having an inner diffusion layer adjacent the chromium-containing layer and an outer additive layer adjacent to the inner diffusion layer. The chromium-containing layer is deposited by a deposition technique that permits chromium in the chromium-containing layer to more readily diffuse into a subsequently deposited aluminde diffusion coating layer. The chromium-containing and aluminide diffusion layers are then treated to cause chromium from the chromium-containing layer to diffuse into the outer additive layer in an amount of at least about 8%. The resulting coated article is resistant to corrosion.

  • Publication number: 20060019292
Abstract: Methods are provided for producing a vector that includes at least one splicable intron. In the subject methods, intron containing vectors are produced from donor and acceptor vectors that each include a site specific recombinase site, where the subject donor and acceptor vectors further include splice donor and acceptor sites that, upon site specific recombination of the donor and acceptor vectors, define an intron in the product vector of the recombination step. Also provided are compositions for use in practicing the subject methods, including the donor and acceptor vectors themselves, as well as systems and kits that include the same. The subject invention finds use in a variety of different applications, including the production of expression vectors that encode C-terminal tagged fusion proteins, the production of expression vectors that encode pure protein and not a fusion thereof, and the like.

  • Publication number: 20050060771
Abstract: siRNA encoding constructs and methods for using the same are provided. The subject constructs are characterized by including a siRNA coding domain flanked by opposing promoters. In using the subject constructs, sense and antisense strands of the desired siRNA product encoded by the coding domain are transcribed under the direction of the two opposing promoters flanking the coding domain. The transcribed sense and antisense strands are then annealed to each other to produce the desired siRNA double-stranded molecule. The subject constructs and methods find use in a variety of applications, including applications where the specific reduction or silencing of a gene is desired. Also provided are systems and kits for use in practicing the subject invention.
