    Abstract: An improved method of making a series of bead or microsphere or particle populations characterized by subtle variation in a proportion or ratio of at least two fluorescent dyes distributed within a single bead of each population is provided. These beads, when excited by a single excitation light source are capable of giving off several fluorescent signals simultaneously. A set containing as many as 64 distinct populations of multicolored, fluorescent beads is provided and when combined with analytical reagents bound to the surface of such beads is extremely useful for multiplexed analysis of a plurality of analytes in a single sample. Thus, methods of staining polymeric particles, the particles themselves, and methods of using such particles are claimed.

Flow cytometric measurements are used to classify, in real-time, beads within an exposed beadset and textual explanations, based on the accumulated data obtained during real-time analysis, are generated for the user. A secondary reagent, such as a metal or magnetic particle, is added to the beadset to assist in the analysis. Detection techniques, such as such as light scatter, Rayleigh scatter, Raman scatter, surface plasmon resonance, magnetic induction, or magnetoresistance are used to detect the particle labels.

    Abstract: Various computer-implemented methods and systems are provided. One computer-implemented method includes determining a ratio between output signals generated by detecting spectra for a single event in two or more detection windows. The spectra are characteristic of different materials. At least a portion of the spectra overlap in at least one of the two or more detection windows. The method also includes determining which of the different materials are associated with the ratio. One embodiment of a system includes one or more detectors configured to detect spectra for a single event in two or more detection windows. The spectra may include spectra as described above. The one or more detectors are also configured to generate output signals in response to the detected spectra. The system also includes a processor configured to determine a ratio between the output signals and to determine which of the different materials are associated with the ratio.
