  • Patent number: 5808047
Abstract: A process for the preparation of compounds of the formula: ##STR1## wherein Pg and R are protecting groups and B is a nucleobase. The process uses protected nucleobases to form 2,2'-anhydro nucleosides having the formula ##STR2## which are hydrolyzed to 2'-hydroxy intermediates which are then oxidized to the desired 2'-keto compounds. The compounds and intermediates are useful as antiviral and antitumor agents and as intermediates to 2,2'-anhydro-1-(.beta.-D-arabinofuranosyl)cytosine (anhydro-ara-C) and 1-.beta.-D-arabinofuranosylcytosine (Ara-C).
    Type: Grant

  • Patent number: 5750688
Abstract: Novel processes are provided for producing compounds of formula II ##STR1## wherein R.sup.1 and R.sup.2 each are independently C.sub.1 -C.sub.4 alkyl, or combine to form piperidinyl, pyrrolidinyl, methylpyrrolidinyl, dimethylpyrrolidinyl, morpholino, dimethylamino, diethylamino, or 1-hexamethyleneimino;R.sup.3 and R.sup.4 each are H or a hydroxy protecting group; andn is 2 or 3;or a pharmaceutically acceptable salt thereof by reacting a haloalkylamine with a phenolic ester in the presence of an alkylacetate solvent and a base.

  • Patent number: 5633367
Abstract: A process for the preparation of a 2-substituted-3,3-difluorofuran using diethyl ammonium sulfur trifluoride (DAST) and pyridine hydrogen fluoride. The process is particularly useful for producing a 2',2'-difluoronucleoside, particularly 1-(2',2'-difluoro-.beta.-D-arabinofuranosyl)cytosine (also known as 2'-deoxy-2',2'-difluorocytidine), a known antiviral and antitumor agent.
