    Abstract: The present invention features methods for measuring the activity of an enzyme (such as a proteinase or an endosaccharidase) in a sample, using a macromolecular substrate of the enzyme. Also featured are methods for: detecting the level of peptidase activity of a proteinase; measuring amylase activity in a sample; diagnosing pancreatitis in a subject; measuring the activity of a target isoenzyme in a sample; identifying a compound that modulates the activity of a proteinase or an endosaccharidase; and identifying an antibody that modulates the activity of a proteinase or an endosaccharidase, using the macromolecular substrates provided by the present invention.
