    Abstract: A method is provided of measuring in vivo of an endogenous fluorophore in a tissue site. A known excitation wavelength of the endogenous flurophore is selected within a range of wavelengths at which the endogenous flurophore undergoes fluorescence. The tissue site is irradiated with irradiated light having at least the selected excitation wavelength within the range of wavelengths. A fluorescence emission of the tissue site resulting from the irradiation thereof is detected. A relative or absolute concentration of the endogenous fluorophore is determined by multiplying it by a calibration factor that depends one at least one of, a known excitation and emission property of the endogenous fluorophore, an intensity of the irradiated light, optical properties of an excitation probe, and specific properties of the tissue. The relative or absolute concentration of the endogenous fluorphore is used to estimate at least one of a, in vivo cellular energy production status or state of end-organ tissue oxygenation.

    Abstract: A method and device for assaying sperm motility in a forward direction and density of active sperm in a semen sample are disclosed. The device includes a microfluidics structure having a sample reservoir, a downstream collection region and a microchannel extending therebetween. The microchannel is dimensioned to confine sample sperm to single-direction movement within the channel, such that sperm in a semen sample placed in the sample reservoir enter and migrate along the microchannel toward and into the collection region. Also included is a detector for detecting the presence of labeled sperm in the microchannel or collection region, and an electronics unit operatively connected to the detector for (i) receiving detector signals, (ii) based on the detector signals received, determining sperm motility and density in the sperm sample, and (iii) displaying information related to sperm motility and density.

The optical module is integrally formed and composed of an upstream portion having a focusing optical surface for directing excitation light from the light source to a focal region within an analyte-detection zone in the apparatus, and a downstream portion having a second focusing powered optical surface and at least one reflecting surface for directing fluorescence-emission light produced by fluorescence events within the detection zone, in a direction substantially normal to the path of the excitation light onto the photodetector. The optical module is adaptable to a variety of assay formats, including multiple-sample, sample-array, and disposable-cartridge formats.
