Publication number: 20020112723Abstract: A portable air temperature controlling device useful for warming air surrounding an aerosolized drug formulation. Warming the air of an aerosol makes it possible to reduce the diameter of aerosol particles produced by an aerosol generation device. Additionally, warming the air forces the diameter of the aerosol particles to be in the range required for systemic drug delivery independent of ambient conditions. Smaller particles can be more precisely targeted to different areas of the respiratory tract.Type: ApplicationFiled: September 20, 2001Publication date: August 22, 2002Inventors: Jeffrey A. Schuster, Joan Rosell, Avi Eliahu, Christopher J. Flaim

Publication number: 20020106331Abstract: Formulations are disclosed as are aerosols created therefrom. The formulations are comprised of (a) a pharmaceutically active drug which does not ionize in solution; (b) an electrolyte; and (c) a solvent which is preferably water and/or ethanol. The electrolyte reduces electrostatic charging on particles of aerosol formed thereby enhancing characteristics of the aerosol particles which are important for efficient, repeatable intrapulmonary drug delivery.Type: ApplicationFiled: December 8, 2000Publication date: August 8, 2002Inventors: Joan Rosell, Igor Gonda, Jeffrey Schuster, Kui Liu

Publication number: 20010010224Abstract: A nozzle comprised of a thin, flexible membrane material having a plurality of pores is disclosed. In one embodiment, the pores have an unflexed exit aperture diameter in the range of about 0.5 to about 2 microns (preferably about 1 micron) and are positioned substantially uniformly in the material, preferably about 50 microns apart. The nozzle preferably has a conical or trumpet-shaped cross-section. In another aspect of the invention, the exit aperture of the nozzle is surrounded by an elevated area protruding above the substantially planar exit side of the membrane in order to prevent intrusion of liquid back into the nozzle. The nozzle can be used to form an aerosol containing a pharmaceutical composition from the exit side of the nozzle upon forcible application of the composition to the entrance side of the nozzle. This aerosol can be used to administer the pharmaceutical composition, for example, to the eye or to a selected portion of the respiratory tract.Type: ApplicationFiled: March 2, 2001Publication date: August 2, 2001Inventors: Igor Gonda, Jeffrey A. Schuster, Rajesh S. Patel

Patent number: 6098620Abstract: Devices, packaging and methodology for efficiently and repeatably creating aerosolized bursts of an analgesic (e.g., narcotic) containing formulation are disclosed. Devices are hand-held, self-contained units which are automatically actuated at the same release point in a patient's inspiratory flow cycle. The release point is automatically determined either mechanically or, more preferably calculated by a microprocessor which receives data from a sensor making it possible to determine inspiratory flow rate and inspiratory volume. The device is loaded with a cassette comprised of an outer housing which holds a package of individual disposable collapsible containers of an analgesic containing formulation for systemic delivery. Actuation of the device forces analgesic formulation through a porous membrane of the container which membrane has pores having a diameter in the range of about 0.25 to 3.0 microns, preferably 0.25 to 1.5 microns.Type: GrantFiled: October 27, 1995Date of Patent: August 8, 2000Assignee: Aradigm CorporationInventors: Lester J. Patent number: 5960792Abstract: A method of treating human patients is provided by the intrapulmonary delivery of a pharmaceutically active peptide formulation. The formulation is automatically released in an aerosolized form from a hand-held, self-contained, portable device comprised of a means for automatically releasing a measured amount of drug into the inspiratory flow path of a patient in response to information obtained from a means for measuring the inspiratory flow rate and determining the inspiratory volume of a patient. Reproducible dosing is obtained by providing for automatic release at the same inspiratory flow rate and inspiratory volume each time drug is released. The device includes a timer to enable a patient to take a drug at the same time each day. Further, overadministration of hormone formulations is avoided by providing a pre-programmed microprocessor designed to avoid overdosing.Type: GrantFiled: October 27, 1995Date of Patent: October 5, 1999Assignee: Aradigm CorporationInventors: Lester J. Patent number: 5906202Abstract: Methodology and devices for delivering aerosolized formulation to target areas of a patient's respiratory tract are disclosed. The device is a hand-held, self-contained unit which is readily portable and capable of measuring a variety of parameters including the patient's total respiratory tract capacity, inspiratory flow rate and inspiratory volume. The device is loaded with a container which includes a drug formulation in a liquid form which container includes an opening which is covered, at least in part, by a porous membrane. The pore sizes are designed so as to provide aerosolized particle sizes which are tailored in size for delivery to the specific target area of the respiratory tract. The device can allow the patient to inhale a predetermined volume of unaerosolized air followed by a predetermined volume of aerosol after which flow can be shut off completely or followed by additional aerosol free air.Type: GrantFiled: November 21, 1996Date of Patent: May 25, 1999Assignee: Aradigm CorporationInventors: Jeffrey A. Patent number: 5770152Abstract: A device and method for the detection of small particles in a fluid. The device is a collapsible sample container having a opening which is covered, at least in part, by a porous membrane having a plurality (e.g., 100) of pores therein having a pore diameter equal to or smaller than the diameter of the particles to be detected (e.g., 01. to 100 microns). A means for collapsing the container to extrude the liquid sample through the membrane pores is provided along with a means for detecting pressure during the extrusion. The size and number of particles in the liquid is calculated by relating the extrusion pressure required to a pre-calibrated standard. When a constant pressure is applied the rate at which fluid is extruded decreases as the pores of the membrane become clogged with particles.Type: GrantFiled: November 18, 1996Date of Patent: June 23, 1998Assignee: Aradigm CorporationInventors: Jeffrey A. Patent number: 5544646Abstract: A disposable package, tape, and cassette are provided which makes it possible to hold and disperse therefrom liquid, flowable formulations including aqueous formulations (solutions or dispersions with particles less than 0.25 microns in diameter) of a pharmaceutically active drug. In one embodiment formulation is packaged in individual dosage unit containers which containers are preferably interconnected. The package is designed to be integrated into a cassette which can be loaded into a dispersing device capable of individually opening dosage unit containers and aerosolizing the contents through a porous membrane, into a mouth piece on the cassette, for delivery to a patient. In addition to and alongside of each porous membrane, the package may include one or more openings through which air is forced in order to aid in avoiding the accumulation of aerosolized particles.Type: GrantFiled: May 20, 1994Date of Patent: August 13, 1996Assignee: Aradigm CorporationInventors: Lester J. Patent number: 5522385Abstract: A device is provided which creates aerosolized particles by forcing a formulation of drug in a carrier through a nozzle (which nozzle may be in the form of a porous membrane) into a channel to a patient for inhalation. While in the channel energy is added to the particles in an amount sufficient to evaporate carrier and thereby reduce particle size. The amount of energy added can be adjusted depending on factors such as the particle size, the amount of the carrier to be evaporated, the water vapor content of the surrounding air and the composition of the carrier. Energy may be added in an amount sufficient to evaporate all carrier and thereby provide particles of dry powdered drug to patient which particles are uniform in size regardless of the surrounding humidity and smaller due to the evaporation of the carrier.Type: GrantFiled: September 27, 1994Date of Patent: June 4, 1996Assignee: Aradigm CorporationInventors: Lester J. Lloyd, Peter M. Lloyd, Reid M. Rubsamen, Jeffrey A. Schuster
