    Abstract: A method for preparing fluorescent-encoded microspheres coated with metal nanoshells is disclosed herein. By using SPG method, metal nano-material modified with a certain ligand is used as a new surfactant in the emulsification process, and different kinds and different amounts of fluorescent materials are doped into polymer microspheres to prepare fluorescent-encoded microspheres with different fluorescent-encoded signals and uniformly coated metal nanoshells in one step. The prepared fluorescent-encoded microsphere comprises a metal nanoshell, a polymer, and a fluorescent-encoded material. The fluorescent-encoded microsphere has a particle size of 1 ?m˜20 ?m, CV of less than 10%, which can be used for protein/nucleic acid detection.

The light blocking element separates photodetectors in separate spaces, including a first light blocking element and a second light blocking element, wherein the first light blocking element is located between the first light-emitting element and the first photodetector, to guide the light emitted from the light emitting element to illuminate the testing element.

The detection method using suspension liquid biochip comprises the following steps: an excitation step and a signal acquisition step. In the excitation step, a laser light emitted by a single laser device is used to excite a classification fluorescence of nanocrystalline fluorescent microspheres and a report fluorescence of an object to be detected; and in the signal acquisition step, fluorescence detection signals are obtained through multiple acquisition channels, respectively. In the present invention, the classification fluorescence in the nanocrystalline fluorescent microspheres and the report fluorescence in the object to be detected in a sample are excited by a single laser device, which is convenient for operation and reduces the production cost of the system, thereby reducing the detection cost.

Moreover, each lysis tube can be inserted into respective packaging box; and each packaging box is corresponding to respective detecting apparatus. Therefore, it is not easy to cause the misuse of lysis tubes during loading and detection procedure, thereby avoiding wrong detection because the lysis tube is taken by mistake.
