Publication number: 20160146786Abstract: This disclosure provides a method of isolating peptides having cell-penetrating function, wherein the peptides are detected as biotinylated molecules only following their translocation through the cell membrane. The disclosure also provides methods for validating the cell-penetrating function of the peptides, or that may be employed in their own right to isolate such peptides, wherein the peptides are detectable by virtue of their ability to transport a detectable cargo into the cytoplasm, such as a cargo toxin or a fragment of a green fluorescent protein (GFP) that is required for complementation of a functional GFP. The disclosure also provides non-canonical peptides having cell-penetrating function that differ structurally from known CPPs such as TAT, VP22, transportan and penetratin, and that are capable of translocating cell membranes and escaping the endosome. The disclosed peptides have utility in transporting cargo therapeutics and diagnostics into cells.Type: ApplicationFiled: June 26, 2014Publication date: May 26, 2016Inventors: Richard Hopkins, Katrin Hoffmann, Tatjana Heinrich, Paula Cunningham, Paul Watt, Nadia Milech

Publication number: 20130266605Abstract: The present invention provides compositions comprising peptidyl inhibitors of CD40L-dependent signaling that are not derived from a natural binding partner of CD40L such as CD40, or from a native CD40-CD40L interface. More particularly, the peptidyl inhibitors of the present invention are derived from natural sources that do not express CD40-Cd40L costimulatory pathways. The invention also provides synthetic derivatives and analogs of the peptidyl inhibitors having enhanced binding affinity for CD40L or enhanced inhibitory activity relative to their parent molecules.Type: ApplicationFiled: September 16, 2011Publication date: October 10, 2013Applicant: PHYLOGICA LIMITEDInventors: Paul Michael Watt, Richard Hopkins, Katrin Hoffmann

Patent number: 7947502Abstract: The invention is directed to a method and a kit for calibrating a photoluminescence measurement system, in particular a fluorescence measurement system. The kit includes a number of fluorescence standards i and their corrected and certified fluorescence spectra Ii(?), whereby the fluorescence standards i are selected, so that their spectrally corrected fluorescence spectra Ii(?) cover a broad spectral range with high intensity. The standards are characterized by large half-widths FWHMi of their bands of at least 1400 cm?1. According to the method of the invention, partial correction functions Fi(?) are generated by forming the quotient of the measured fluorescence spectra Ji(?) and the corresponding corrected fluorescence spectra Ii(?), which are then combined to form a total correction function F(?) for a broad spectral range.Type: GrantFiled: March 23, 2010Date of Patent: May 24, 2011Assignees: Sigma-Aldrich GmbH, BAM Bundesanstalt für Materialforschung und-prüefungInventors: Ute Resch-Genger, Dietmar Pfeifer, Christian Monte, Angelika Hoffmann, Pierre Nording, Bernhard Schönenberger, Katrin Hoffmann, Monika Spieles, Knut Rurack
