    Abstract: A lytic reagent composition and the method of use for differential analysis of leukocytes using flow cytometry are disclosed. The lytic reagent composition includes a short chain alkyl oxyethanol, such as 2-methoxyethanol, 2-ethoxyethanol, 2-propoxyethanol, or 2-isopropoxyethanol, in a sufficient amount to preserve leukocytes; a non-lysing nonionic surfactant as a debris solublizer; and an inorganic buffer to maintain pH of the lytic reagent composition in a range from 9.1 to 10.7. The lytic reagent composition is used to lyse red blood cells and preserve leukocytes, as well as used as the sheath reagent for the focus flow measurement. The lytic reagent composition, when used on a flow cytometric analyzer with multiple angle light scatter and light absorbance measurements, enables differentiation of leukocytes into five subpopulations.

    Abstract: The present invention relates to a hematology control product for automated hematology instruments containing an aqueous solution of at least one blood cell analog, an absorbance agent in an amount sufficient to simulate a hemoglobin concentration, and a stabilizing agent in an amount sufficient to stabilize the size of the blood cell analog when the control product is subjected to temperature ranging from -15.degree. C. to 45.degree. C. The control product does not require controlled temperature storage for product stability. In addition, a novel method of using a hematology control product is provided wherein the control product contains a single blood cell analog and is used to simulate both red blood cells and white blood cells on an automated hematology instrument.

    Abstract: A hematology control product comprising leukocyte analogs and an aqueous solution of a plasma substance for use in a blood counting instrument is described. The instrument employs a lytic reagent system for the lysable red blood cells in the control product. The plasma substance is in an amount effective to enable the differentiation of each of said leukocyte analogs relative to the physical attributes of the analogs, said physical attributes of the analog are similar to human leukocytes. Preferably, the plasma substance comprises cholesterol or its derivatives.A method for using the cell suspension media comprising an aqueous solution of a plasma substance is also described. The method provides a quality control to determine whether an instrument is operating within manufacturer's specifications.

A further method is described to use the control product to determine whether an automatic instrument is operating within manufacturer's specification.
