    Abstract: The present invention provides a Ganoderma lucidum ?-glucan extract, a preparation method and detection method therefor. The preparation method includes: S1: crushing fruiting bodies of Ganoderma lucidum, mixing the crushed fruiting bodies with a solvent, adding trypsin for enzymatic hydrolysis for 0.5-2 h, and separating to obtain a Ganoderma lucidum fruiting body filter residue; S2: mixing the Ganoderma lucidum fruiting body filter residue with an alkali solution, carrying out ultrasonic extraction with heating, adjusting the pH value of the obtained filtrate to be neutral, and concentrating to obtain a crude Ganoderma lucidum polysaccharide; S3: carrying out alcohol precipitation on the crude Ganoderma lucidum polysaccharide, separating the obtained precipitate, and freeze-drying to obtain a crude Ganoderma lucidum ?-glucan extract; and S4: purifying the crude Ganoderma lucidum ?-glucan extract by glucose gel column chromatography to obtain a Ganoderma lucidum ?-glucan extract.

The present disclosure uses Saccharomyces cerevisiae CEN.PK2-1D as a host, and implements knockout of ROX1 and GAL80 genes on the genome, and integrative expression of GGPP synthase encoding gene and CPS diterpene synthase encoding gene at ROX1 site; and implements free expression of ApCPR and CYP71A8 and CYP71D10 both with truncated signal peptides, successfully constructing recombinant S. cerevisiae, and achieving de novo synthesis of 3,15,19-Trihydroxy-8(17),13-ent-labdadiene-16-oic acid. Compared with the blank, a response value of a product peak reaches 1.9*106, and this strategy provides necessary reference for analyzing biosynthetic pathway of andrographolide and using metabolic engineering to synthesize andrographolide and related derivatives thereof.

This blending model is applied to a combination of both sponsored and non-sponsored pieces of content being considered for display to a user. This pattern only determines the slot assignments. The actual ranking of the pieces of content, and more particularly the actual ranking of the organic pieces of content, is determined by an ordering other than the ranking determined by the blending model, such as by using the original ordering of the second list. The pieces of content are then displayed in the order of this actual ranking, but in the slots indicated as having been assigned to be either sponsored or organic in the pattern determined by the blending model.

    Abstract: In an example embodiment, a machine-learned model is trained to forecast a performance-based metric for a piece of content based on a budget applied to the piece of content. The increase in the performance-based metric that is due to a corresponding increase in budget may be termed “incrementality.” The machine-learned model is trained in such a way that incrementality is built into the model. More particularly, in an example embodiment, an asymmetric budget split process is used to create two groups of training data, one for high budget and one for low budget. Rather than relying on historical data, the asymmetric budget split process applies a high budget to a piece of content in a first subgroup (e.g., group of users) and a low budget to that same piece of content in a second subgroup, and then the performance results in each subgroup are compared.
