    Abstract: A method for detecting the presence of an analyte in a solution. The analyte includes at least two mutually exclusive recognition sites that are capable of binding to corresponding recognition molecules. Biosensors are provided that include the recognition molecules, which are attached to the inactive portions of a split enzyme. The recognition sites are located such that the inactive enzyme portions combine to form a detectable biologically active enzyme when the recognition molecules bind to recognition sites.

    Abstract: Fluorescence, spectroscopy is used to analyze small numbers of molecules that are present in a relatively small detection volume or zone. Information regarding physical and chemical properties of these molecules is determined by rapidly modulating the wavelength, intensity and/or polarization of laser energy to excite fluorophores that are attached either to the molecule of interest or a molecule that interacts with the molecule of interest. The emission profile of the fluorophores is used to determine useful information about the labeled and/or non-labeled molecules including molecular interactions between th molecules.

    Abstract: A method for analyzing/monitoring the properties of species that are labeled with fluorophores. A detector is used to detect photons emitted from species that are labeled with one or more fluorophores and located in a confocal detection volume. The arrival time of each of the photons is determined. The interval of time between various photon pairs is then determined to provide photon pair intervals. The number of photons that have arrival times within the photon pair intervals is also determined. The photon pair intervals are then used in combination with the corresponding counts of intervening photons to analyze properties and interactions of the molecules including brightness, concentration, coincidence and transit time. The method can be used for analyzing single photon streams and multiple photon streams.

Two unique families of fluorophores are used, namely energy-transfer fluorescent beads and semiconductor nanocrystal (NC) quantum dots, that can be excited by a single laser wavelength but emit at different wavelengths. A novel multicolor sample-scanning confocal microscope was constructed which allows one to image each fluorescent light emitter, free of chromatic aberrations, by scanning the sample with nanometer scale steps using a piezo-scanner. The resulting spots are accurately localized by fitting them to the known shape of the excitation point-spread-function of the microscope.
