  • Publication number: 20210403942
Abstract: The disclosure relates to growing cells, directing cells to grow into specified cell types, genetically and physically manipulating cells, and addressing one or more individual cells within a mixed cell population. Aspects of the disclosure relate to vectors useful to induce developmental changes in cells, in which those vectors have a temporal component. Vectors of the disclosure encode a controllable, temporal series of events. Once the vectors are delivered into target cells, a series of discrete and different genetic events may be induced. The disclosed methods generally provide for the temporal encoding of multiplex genetic effectors in vector format for cell state transitions.

  • Publication number: 20210301247
Abstract: Systems and methods for image-driven cell manufacturing are provided. Systems comprise a substrate for cells, imaging system for imaging cells on the substrate, computing system that computes cell characteristics from images, and a pulsed laser scanning system. The substrate is suitable for high-resolution cell imaging and is coated with a layer that partially absorbs laser pulses for the purpose of converting energy into microbubble formation. The computing system communicatively coupled to the pulsed laser scanning system and directs laser pulses to the substrate under targeted cells. Laser pulses are converted into mechanical energy via microbubbles that, depending on laser energy and pulse pattern, destroy selected cells, remove selected cells, or temporarily porate selected cells for the purpose of introducing biological cargos.

  • Publication number: 20210254049
Abstract: Methods for identifying targets involved in cell differentiation, for instance particular loci of a human genome. To identify such targets, for example, complexes that each include a catalytically-deactivated DNA binding protein, a guide RNA that guides the complex, and one or more effector domains may be introduced into stem cells to cause at least one of the stem cells to differentiate into a target phenotype. The guide RNAs present in cells demonstrating the target phenotype may be identified and a nucleic acid sequence of each identified guide RNA may be correlated to loci of a genome to identify targets involved in directing cell differentiation to the target phenotype. These methods may be used for directed cell fate specification in stem cells, such as induced pluripotent stem cells, to produce synthetic cells with a desired target phenotype.

  • Patent number: 7686629
Abstract: An electronic device is provided for connecting to an external device. The electronic device includes a main body, a connector at a proximal end of the main body, a bottom case coupled to the main body at a distal end, and a top case slidably located opposite to the bottom case. The electronic device further includes a first cantilever locking arm and a second cantilever retaining arm on at least one side of the main body. The top case is slid towards the bottom case to expose the connector and the top case is slid towards the connector to cover the connector. The first cantilever locking arm includes an open locking mechanism and a close locking mechanism for maintaining the top case in an open state and a closed state respectively. The second cantilever retaining arm retains the top case on the main body.
