  • Publication number: 20150044666
Abstract: Methods and kits related to non-equilibrium, ultrasensitive two-site assays for detecting analytes are provided. In one aspect, a two-site assays for detecting analytes under non-equilibrium analyte binding conditions, using low concentrations of reporter specificity molecule (e.g., reporter antibody) and kits for performing the same is provided. In another aspect, methods for selecting antibodies or specificity molecules with low dissociation constants for use as reporter antibodies in non-equilibrium two-site immunoassays, including two-site immuno-PCR assays, and assays performed with those antibodies, are also provided.

  • Patent number: 5710029
Abstract: Methods and kits for relating initial amounts of target nucleic acids present in a sample to target-specific amplification products. It has been discovered that the transcription-mediated amplification system is capable of producing a quantitative relationship between target input and target-specific output. Further, the present invention relates to methods for carefully controlling this relationship resulting in an unexpectedly high degree of reproducability. Also described are useful methods for extending the dynamic range of transcription-based amplification systems.
    Type: Grant
    Filed: June 7, 1995
Date of Patent: January 20, 1998
    Assignee: Gen-Probe Incorporated
Inventors: Thomas Brendan Ryder, Karen W. Shannon, Daniel Louis Kacian, Richard C. Harvey, Sherrol H. McDonough, Frank R. Gonzales, Maria R. Castillo, Elizabeth R. Billyard, Nancy Lau Liu Shen
