Abstract: Methods of making orally available softgels includes pre-wetting a probiotic with an edible oil to produce a wetted supplement, wherein the edible oil is present in a range of 25% to 75% by weight of a total weight of the probiotic and edible oil, heating the wetted supplement to a temperature in a range of 35° C. to 40° C., blending the wetted supplement and a molten chocolate component together to produce a liquid fill material, encapsulating the liquid fill material in a softgel coating to produce an orally available softgel, and cooling the orally available softgel to have a softgel capsule shell comprising 5% to 15% by weight water. The fill material is a solid at room temperature in the cooled orally available softgel.

Abstract: Softgel capsules have a fill composition that administers a 200 to 500 mg dose of magnesium from magnesium oxide and an outer sofgel gelatin shell. The composition includes 55 to 70% w/w of an edible oil, 2 to 8% w/w polysorbate 80, 0.5 to 2% w/w hydrophobic silica, and 0.2 to 4% w/w of a chelating agent for binding to magnesium. The softgel capsules have a dissolution according to USP <711> of greater than NLT 75% (Q).

Abstract: Film-forming compositions for soft capsule formation have from 2 to 20 percent by weight of a mixture of hydroxyalkylated plant starch and a plant sugar, from 15 to 25 percent by weight agar, no more than 5 percent by weight gelatin, no more than 1 percent by weight carrageenan, and water as the balance thereof. Soft capsules made from such a film-forming composition and methods of making the same are also disclosed.
