The methods and compositions involve increasing, promoting, restoring, and/or enhancing differentiation of oligodendrocyte progenitor cells, myelin protein expression, mature oligodendrocyte marker expression, and/or myelination. The methods and compositions disclosed herein inhibit or block pathological activation of RhoA in OPCs.
