    Abstract: In the present invention, a method of producing stable bare colloidal gold nanoparticles is disclosed. The nanoparticles can subsequently be subjected to partial or full surface modification. The method comprises preparation of colloidal gold nanoparticles in a liquid by employing a top-down nanofabrication method using bulk gold as a source material. The surface modification of these nanoparticles is carried out by adding one or multiple types of ligands each containing functional groups which exhibit affinity for gold nanoparticle surfaces to produce the conjugates. Because of the high efficiency and excellent stability of the nanoparticles produced by this method, the fabricated gold nanoparticle conjugates can have surface coverage with functional ligands which can be tuned to be any percent value between 0 and 100%.

    Abstract: Disclosed is a method and a kit for multi-color cell imaging with dark field optical microscopy using noble metal conjugated nanoparticles. The noble metal conjugated nanoparticles include a stabilizer component and a binding ligand, the stabilizer component coats a portion of the noble metal nanoparticle keeping it stable in biological buffers and cell cytoplasm. The binding ligand specifically binds to targeted cells designated for imaging. The method and kit permit multicolor imaging of cells, with the multiple colors being derived from localized surface plasmon resonance of the nanoparticles, each color the result of different amounts of one or more noble metals in the nanoparticle.

The portion of the base that is removed can be removed such that a recess is defined in the base and/or a shelf is defined on the device precursor. A light source such as an optical fiber or laser can be received in the recess and/or positioned over the shelf such that the light source is optically aligned with the facet of the waveguide.
