    Abstract: Disclosed is a tumor vaccine based on tumor-derived microparticles (TMPs), and a preparation method and use thereof. The preparation method includes the following steps: 1, subjecting tumor cells to separation to obtain TMPs; 2, mixing the TMPs with 0.03 ?g/?l of a polyethyleneimine solution to allow a reaction for 10 min to 20 min, and washing to obtain PEI-modified TMPs (TMPs-PEI); and 3, mixing the TMP-PEI with 0.1 ?g/?l of a lipopolysaccharide solution to allow cross-linking for 15 min to 1 h to obtain the tumor vaccine based on TMPs (TMP-PEI-LPS). A nanomaterial prepared by conducting surface modification on the TMPs with an immunologic adjuvant can be disguised as a bacterial analog to promote the recognition of tumor antigens in vivo. Moreover, the TMPs can retain their immunostimulatory ability and reduce their toxicity by using a cross-linking agent coupled with the immunologic adjuvant.

    Abstract: A method for preparing a tumor-derived microparticle by a microwave includes the following steps: step 1, Lewis lung carcinoma (LLC) of a lung adenocarcinoma cell line is taken and then the LCC is cultured in a culture dish for more than 24 hours (h) to obtain cultured cells; step 2, microwave heating treatment is performed on the cultured cells obtained in step 1 to obtain treated cells; step 3, the treated cells obtained in step 2 are placed into a constant temperature incubator for cultivation for 24 h; and step 4, a supernatant of cells is collected from the culture dish cultured in step 3, and multiple centrifugation treatments is performed on the supernatant by using a density gradient centrifugation method to obtain a precipitate which is the tumor-derived microparticle TMPMW.

    Abstract: A preparation method of a tumor microparticle encapsulating a metabolic inhibitor includes following steps. A tumor cell is cultivated, and a statin drug is added to a culture medium after the tumor cell grows stably. Then, the tumor cell is irradiated with an ultraviolet, and then the tumor cell is incubated in a cell incubator for 22˜26 hours. After the incubation, cell supernatant is collected and the tumor microparticle encapsulating the metabolic inhibitor is obtained through a gradient centrifugation. The preparation method uses tumor cell-derived microparticles as drug delivery platforms, with a simple preparation process that preserves functions such as biosafety, biocompatibility, targeting, and intercellular communication. The preparation method discovers new uses of a statin drug, which can inhibit tumor growths with the statin drug, regulate tumors metabolism and improve a tumor microenvironment, and have a potential to enhance a chemotherapy efficacy and delay a chemotherapy resistance.
