    Abstract: The present invention provides modified promoters from Candida troplicalis CYP and POX4 genes. The modified promoters have various sequence motifs added, deleted, or altered in order to modulate expression of a coding sequence operably linked thereto. The sequence motifs comprise repressors of gene induction (URS sequences) and activators of gene induction (UAS sequences) as well as oleic acid response elements (ORE sequences). Yeast host cells comprising such modified promoters are also provided. Methods of altering expression of a protein of the beta or omega oxidation pathways using a subject modified promoter are also provided.

    Abstract: The present invention provides antigenic peptides useful for the production of antibodies which selectively bind to an enzyme involved in the ?-oxidation of fatty acids and alkanes to ?,?-dicarboxylic acids in yeast. Antibodies which specifically bind to an enzyme involved in the ?-oxidation of fatty acids and alkanes to ?,?-dicarboxylic acids in yeast are also provided. In addition, methods of producing the subject antibodies, a method of detecting the presence and amount of a specific enzyme involved in the ?-oxidation of fatty acids and alkanes to ?,?-dicarboxylic acids, and a method of monitoring the degree of enzyme induction and/or enzyme stability in a mixture during ?-oxidation of fatty acids and alkanes to ?,?-dicarboxylic acids in yeast, are also provided.

    Abstract: The present invention provides methods of converting or increasing conversion of a fatty acid to its corresponding dicarboxylic acid. The methods comprise isolating a promoter from a yeast gene which gene is induced when the yeast is grown on a fatty acid or alkane substrate, and operably linking the promoter to a gene involved in dicarboxylic acid production to form an expression vector. Yeast cells are subsequently transformed with such an expression vector and cultured in a media containing an organic substrate biooxidizable to a mono- or polycarboxylic acid, and resultant yeast cells convert or increase conversion of fatty acids to their corresponding dicarboxylic acids. Examples of promoters that may be used in the methods of the present invention include those from C. tropicalis catalase, citrate synthase, 3-ketoacyl-CoA thiolase A, citrate synthase, O-acetylhomserine sulphydrylase, protease, carnitine O-acetyltransferase, hydratase-dehydrogenase, and epimerase genes.
