    Abstract: The present disclosure provides a kit and method for constructing a long-acting depression animal model. The present disclosure adopts Bacille Calmette-Guerin (BCG) (or Mycobacterium bovis (M bovis)) and low-dose pertussis toxin (PTX) for combined induction, which greatly reduces modeling cost. A mouse model constructed by the method of the present disclosure can clearly distinguish a time interval of pathological behaviors from a time interval of depression-like behaviors and has prominent face validity and predictive validity. The method of the present disclosure is simple and convenient and does not require a large number of tedious daily operations as the depression model construction by chronic unpredictable stress, chronic restraint stress, social defeat, mother-child separation, or the like. The mouse model of the present disclosure can provide a valuable long-term intervention window for research on antidepressant therapy.

    Abstract: The present disclosure provides a successive gas path fault diagnosis method with high precision for gas turbine engines and falls within the technical field of fault diagnosis for gas turbine engines, including the following steps: establishing an engine nonlinear component-level model; capturing dynamic effects of an engine transient maneuver; outputting an estimated value of an engine observation parameter by the engine nonlinear component-level model; acquiring a measurement of the engine observation parameter through sensors; and iteratively updating a degradation factor through a solver. The present disclosure captures the dynamic effects of the transient maneuver at consecutive moments through time-series gas path measurement parameters, thereby realizing successive and high-precision diagnosis for health conditions of the gas turbine engines. This technology can provide a new successive and high-precision diagnosis method for the gas turbine engines under steady-state and transient conditions.

    Abstract: The invention provides herpes virus nucleic acid vectors for expressing shRNAs in mammalian cells and thereby silencing target genes. The vectors include (a) a herpes virus packaging signal sequence; (b) a herpes virus origin of replication; (c) a segment expressing a light-emitting marker; and (d) a transfer RNA promoter upstream of a restriction endonuclease recognition sequence. A segment encoding an shRNA can be cloned into the restriction endonuclease recognition sequence. Thus, the invention also provides vectors containing: (a) a herpes virus packaging signal sequence; (b) a herpes virus origin of replication; (c) a segment expressing a light-emitting marker; and (d) a transfer RNA promoter linked to (e) a segment encoding a short hairpin RNA (shRNA) that is adapted to degrade in vivo to a small interference RNA (siRNA) that is complementary to a segment of a target gene.
