Publication number: 20090088721Abstract: The present invention provides a device and a method for the enhanced retinal delivery of nucleic acid therapeutics utilizing iontophoresis to evoke a transient elongation of the Müller Cells of a mammalian eye. The enhanced retinal deposition can be achieved by either a topical application, subconjunctival, or an intravitreal injection of the nucleic acid composition followed by, preceded by, or administered simultaneously with the iontophoretic application. The present invention thus provides a particularly advantageous method for the treatment of ocular diseases comprising the in vivo administration of a nucleic acid capable of alleviating the symptoms of a disease, the delivery of the nucleic acid being enhanced by using iontophoresis. This method can be applied particularly to the diseases of the retina resulting from an alteration of a gene expression and/or the over-expression of particular growth factors.Type: ApplicationFiled: December 5, 2007Publication date: April 2, 2009Inventors: Therese de Bizemont, Florian Sennlaub, Francine Behar-Cohen, Yves Courtois, Andrieu Charlotte

Gene therapy with chimeric oligonucleotides delivered by a method comprising a step of iontophoresis
Publication number: 20030045830Abstract: The present invention provides a method for enhancing the in vivo delivery of chimeric oligonucleotides, containing for example DNA/2′OMeRNA, into cells of a plant, an animal or a human, comprising a step of applying topically to or injecting into a tissue, or tissue adjacent to a tissue, containing said cells, a composition comprising said chimeric oligonucleotide, followed by, preceded by, or simultaneously to a step of transferring said chimeric oligonucleotide into said cells by iontophoresis, and relates to a gene therapy method comprising the iontophorically transfer of a chimeric oligonucleotide DNA/2′OMeRNA. The present invention is also directed to particular chimeric oligonucleotides DNA/2′OMeRNA capable of inducing or inhibiting the expression of a specific gene involved in eye function by inducing or reverting a mutation in that specific gene, and their use as therapeutic composition for preventing or treating ocular diseases.Type: ApplicationFiled: April 17, 2001Publication date: March 6, 2003Inventors: Therese de Bizemont, Florian Sennlaub, Francine BEHAR-COHEN, Yves Courtois -

Patent number: 4872108Abstract: A priority processor (1) and a non-priority processor (20) cooperatively access a common memory (30) by means of an address multiplexer (40) which memory and multiplexer are controlled by a control unit (60). The priority processor issues data strobe (DSSN), clock (CLK) and write control (WSN) signals to the control unit to which the non-polarity processor also issues various memory access request signals. By forming a preparation signal (DSSN=0), the priority processor, through the control unit, claims the memory for a memory access cycle if a prior memory access request by the non-priority processor occurred less than about a clock cycle earlier.Type: GrantFiled: February 24, 1988Date of Patent: October 3, 1989Assignee: U.S. Philips CorporationInventors: Andre Bussonniere, Yves Courtois

Patent number: 4477435Abstract: A composition and a method for regenerating wounded or damaged corneal epelium are described. The composition comprises an aqueous solution containing an effective amount of ocular extract RE. The RE is extracted from eye tissues, such as choroid, iris and vitreous as well as aqueous humor. The active extract is obtained by the treatment of bovine ocular tissues with a buffered aqueous saline solution at pH of about 7.2. For ophthalmological use, the RE extract is purified by precipitation with acetic acid and dialyzed against physiologic saline NaCl solution. Such a composition is shown to increase significantly the rate of healing of wounded corneal epithelium.Type: GrantFiled: September 30, 1982Date of Patent: October 16, 1984Assignee: Institut National de la Sante et de la Recherche Medicale (INSERM)Inventors: Yves Courtois, Denis Barritault, Yves Pouliquen
