    Abstract: The invention provides a polynucleotide comprising portions of the genomes of caprine arthritis-encephalitis virus and HIV-1, resulting in a chimeric retrovirus referred to as a “CHIV. ” The invention also provides a vaccine comprising a CHIV immunogen and a pharmaceutically acceptable carrier. A method of stimulating an immune response in an individual-against human immunodeficiency virus-1 infection by administering a therapeutically effective amount of a CHIV immunogen is also provided. The invention further provides a method of stimulating an immune response in vitro by contacting a lymphocyte with a therapeutically effective amount of a CHIV immunogen.

Another aspect of the present invention is a method of modifying the functional characteristics of a protein including providing a protein and reacting the protein with a glycosylated methanethiosulfonate reagent under conditions effective to produce a glycoprotein with altered functional characteristics as compared to the protein. In addition, the present invention relates to methods of determining the structure-function relationships of chemically modified mutant proteins.

    Abstract: Imaging hardware, software, calibrants, and methods are provided to visualize and quantitate the amount of Fluorescence Resonance Energy Transfer (FRET) occurring between donor and acceptor molecules in epifluorescence microscopy. The MicroFRET system compensates for overlap among donor, acceptor, and FRET spectra using well characterized fluorescent beads as standards in conjunction with radiometrically calibrated image processing techniques. The MicroFRET system also provides precisely machined epifluorescence cubes to maintain proper image registration as the sample is illuminated at the donor and acceptor excitation wavelengths. Algorithms are described that pseudocolor the image to display pixels exhibiting radiometrically-corrected fluorescence emission from the donor (blue), the acceptor (green) and FRET (red).

    Abstract: A communications earpiece comprises an ear canal tube sized for positioning in an ear canal of a user so that the ear canal is at least partially open for directly receiving ambient sounds. A microphone port in the ear canal tube is located in the ear canal for detecting sounds in the ear canal. A speaker port in the ear canal tube is located in the ear canal in close proximity to the microphone port for broadcasting sound signals into the ear canal. A sound processor amplifies the ambient sounds received by the microphone to produce processed analog signals. The sound processor also comprises noise cancellation means for producing an inverse noise signal of noise detected in the ear canal by the microphone. The inverse noise signal is sent to the speaker port so as to be broadcast into the ear canal thereby substantially canceling the ambient noise in the ear canal.

    Abstract: A low defect (e.g., dislocation and micropipe) density silicon carbide (SiC) is provided as well as an apparatus and method for growing the same. The SiC crystal, growing using sublimation techniques, is preferably divided into two stages of growth. During the first stage of growth, the crystal grows in a normal direction while simultaneously expanding laterally. Although dislocation and other material defects may propagate within the axially grown material, defect propagation and generation in the laterally grown material are substantially reduced, if not altogether eliminated. After the crystal has expanded to the desired diameter, the second stage of growth begins in which lateral growth is suppressed and normal growth is enhanced. A substantially reduced defect density is maintained within the axially grown material that is based on the laterally grown first stage material.

    Abstract: To elucidate molecular mechanisms in learning and memory, the expression of mRNAs in brains of rabbitsundergoing eyeblink conditioning was analyzed. Infusion of the transcription inhibitor actinomycin D (ActD) into the cerebellar nuclei reversibly blocked learning but not performance of the CR. Differential display PCR (DD-PCR) analysis of cerebellar RNAs from trained and pseudo-trained rabbits identified a 207-bp band that was induced with learning. The fragment was used to isolate a cDNA from a &lgr;gt11 rabbit brain library containing a 1698-bp open-reading frame. The genomic sequence also has been obtained and is reported. The deduced amino acid sequence contains the KKIAMRE motif, which is conserved among cdc2-related kinases. These results suggest that there is a new category of cdc2-related kinases in the brain whose function may be important in learning and memory.
