Abstract: The subject invention relates to compositions comprising an enzyme mixture which comprises a first enzyme and a second enzyme, where the first enzyme comprises a DNA polymerization activity and the second enzyme comprises an 5?-3? exonuclease activity and a reduced DNA polymerization activity. The invention also relates to the above compositions in kit format and methods for high fidelity DNA synthesis using the subject compositions of the invention.

Abstract: An apparatus for changing relative concentrations of first and second analyte substances in an analyte sample comprises a sample cell defining a sample chamber therewithin, and a semipermeable boundary member disposed in the sample chamber to define first and second sides of the sample chamber. Sample flow input and sample flow output ducts direct an analyte sample containing respective initial concentrations of the first and second analyte substances into the first side of the sample chamber. The semipermeable boundary member permits diffusion therethrough of the first and second analyte substances to the second side of the sample chamber at different rates. An analyte sample in the second side of the sample chamber, and an analyte sample exiting the first side via the sample flow output duct, have respective concentrations of the first and second analyte substances that are different from the initial concentrations.
Abstract: A method for measuring a property of a binding interaction between a capture agent and a binding partner for the capture agent is provided. In certain embodiments, this method comprises: a) contacting a population of particles that are linked to a capture agent with a substrate comprising a binding partner to produce capture agent/binding partner complexes, wherein the population of particles comprises first particles that are bound to a single molecule of the capture agent and second particles that are bound to two molecules of the capture agent; b) applying a force to the bound support, wherein the force is in a direction that separates the particles from the support; and c) separately measuring the forces required to disassociate the first particles and the second particles from their respective complexes.
Abstract: A method and compositions for sulfurizing at least one phosphite or thiophosphite linkage in an oligonucleotide. The addition of N-alkyl imidazole to the acetyldisulfide sulfurization solution enables the use of industrially preferred solvents or solvents that are derived from renewable resources.

Abstract: An apparatus, system, and process of converting a standard, high performance liquid chromatography (HPLC) flow path to a flow path suitable for supercritical fluid chromatography (SFC) are described. This reversible technique is applied to a variety of flow configurations including binary, high pressure solvent mixing systems and quaternary, low pressure solvent mixing systems than can be conventionally operated or automated. The technique is generally applied to the fields of supercritical fluid chromatography and high pressure liquid chromatography, but users skilled in the art will find utility for any flow system where pressurization components must be periodically applied to and removed from both ends of a flow stream in an automated manner.

Abstract: An aerosol is produced by flowing a liquid sample through a gas-assisted nebulizer. The liquid exits from an outlet into a coaxial flow of gas. The outlet includes a sharp edge that inhibits or prevents accumulation of precipitates from the liquid, thereby reducing or eliminating clogging, which is particularly useful for a samples containing high concentrations of dissolved particles. The aerosol may be introduced into a plasma such that molecules are broken into atoms. The atomization may be followed by an analysis such as by optical emission spectrometry or mass spectrometry.
Abstract: A method of sample analysis is provided. In certain embodiments, the method involves: a) obtaining a fragmented RNA sample comprising fragments of long RNA molecules and short RNA molecules; b) ligating an adaptor to an end of the RNA of the fragmented RNA sample to produce an adaptor-ligated sample; c) hybridizing said adaptor-ligated sample to an array of nucleic acid probes; and d) reading said array to obtain an estimate of the abundance of a long RNA in the RNA sample and an estimate of the abundance a small RNA in the RNA sample.

Abstract: An optical absorption spectrometry system includes first and second light sources, a dichroic beam combiner and a wavelength selective module. The first light source emits first light having first wavelengths within a first wavelength range, and the second light source emits second light having second wavelengths within a second wavelength range different from the first wavelength range. The dichroic beam combiner includes a predetermined first reflectance/transmission transition region, the dichroic beam combiner being configured to transmit a first portion of the first light and to reflect a second portion of the second light to provide combined light. The wavelength selective module is configured to disperse the combined light received at an entrance aperture, to select a sample wavelength range of the dispersed light as sample light, and to output the sample light having the selected sample wavelength range from an exit aperture for illuminating a sample.
Abstract: Solvent extraction to quantify contamination of a sample by a hydrocarbon contaminant is performed by providing a defined quantity of the sample, providing a defined quantity of a siloxane solvent, mixing the defined quantity of the siloxane solvent and the defined quantity of the sample to extract the hydrocarbon contaminant from the sample to form a contaminant solution with the siloxane solvent, and separating the contaminant solution from the sample. A concentration of the hydrocarbon contaminant in the contaminant solution can be measured by vibrational spectroscopy. Siloxane solvents are CFC free, VOC exempt, odorless, colorless, low to moderately flammable, non-toxic, and safe for incidental skin contact. Some are even used in cosmetic products.

Abstract: An apparatus includes an electromagnetic which supports propagation of an electromagnetic wave in a first direction between a first end thereof and a second end thereof, and an electromagnetic-field shaping structure within the electromagnetic waveguide. The electromagnetic-field shaping structure defines a channel extending from a first aperture in a first wall of the apparatus to a second aperture in a second, opposite, wall. The channel has an axis extending in a second direction which is nonparallel with the first direction. The distance between the first aperture and the second aperture in the second direction is less than the width of the interior region of the waveguide at the first and second ends thereof. In some embodiments, a plasma torch is disposed within the channel. The length of the torch closely matches its interaction region.

Abstract: A method for operating a data processing system to determine the operational parameters of a communication system between an REC and an RE that utilizes a CPRI data link for communication. The data processing system is synchronized with the CPRI frames and determines a plurality of CPRI frame characteristics from the CPRI frames by examining a plurality of predetermined locations in the CPRI frames. The plurality of CPRI frame characteristics for one or more of the CPRI frames, and a first model of the communication system are used to generate a first broadcast frame that would be generated by the one or more CPRI frames if the first model accurately described the communication system. The first broadcast frame is tested to determine if the first broadcast frame is consistent with the first model. If the broadcast frame is inconsistent, a new model is chosen and the process repeated.
Abstract: A needle for handling a fluid in an analysis system is described. The needle includes a needle body made of a ceramic material and having a fluid conduit extending between a fitting end and a seat end. The fitting end is configured to be connected to a fitting and the seat end (308) being insertable into a seat. The needle body is tapering towards the fitting end. A fixing body is arranged on the needle body next to the fitting end for exerting an axial force when the needle body is connected to the fitting, and a slide-on element to be slid over the needle body so as to push the fixing body towards the fitting end.

Abstract: An external cavity laser and method for operating same are disclosed. The external cavity laser includes a gain chip, a grating, an actuator and actuator driver. The grating diffracts light leaving the gain chip in a band of wavelengths back to the gain chip, the grating is characterized by an angle relative to light leaving the gain chip and distance from the gain chip, the angle and distance is controlled by the actuator. Part of the diffracted light is amplified by the gain chip. The actuator driver causes the angle to be dithered about an equilibrium angle in a motion characterized by an amplitude and average frequency. The amplitude of the dithering motion is chosen to either excite a plurality of adjacent laser modes of the gain chip or provide a servo signal for maintaining the grating angle at a target value.

Abstract: An ion source is configured for electron ionization and produces coaxial electron and ion beams. The ion source includes an ionization chamber along an axis, a magnet assembly configured for generating an axial magnetic field in the ionization chamber, an electron source, and a lens assembly configured for directing the ion beam out from the ionization chamber along the axis, reflecting the electron beam back toward the electron source, and transmitting higher energy ions out from the ion source while reflecting lower energy ions toward a lens element for neutralization.

Abstract: An apparatus having a variable angle light source characterized by a pivot point, a variable response optical receiver, and a first optical system is disclosed. The variable response optical receiver receives light generated by the light source on a receiving surface, the receiver generating a signal indicative of an intensity of light that impinges on a receiving surface. The first optical system images the pivot point to a fixed point relative to the receiver surface. In one aspect of the invention the first optical system is chosen such that light from the variable angle light source covers mores than half the receiving surface. The variable angle light source can include a gain chip in a semiconductor laser having a pivot point located substantially on a facet of the gain chip.
