    Abstract: Solid samples having a uniform sorbate content are produced by directing a gas having a constant sorbate pressure through a plurality of containers, each container enclosing a solid sample. The sorbate content of the solid samples equilibrates with the sorbate pressure in the gas, thereby causing the solid samples to have a uniform sorbate content. An apparatus that can be employed to generate solid samples having a uniform sorbate content includes a relative humidity generator, and a manifold which connects the relative humidity generator to a plurality of solid sample containers.

    Abstract: Injectable compositions having improved injectability. The injectable compositions include microparticles suspended in an aqueous injection vehicle having a viscosity of at least 20 cp at 20° C. The increased viscosity of the injection vehicle that constitutes the fluid phase of the suspension significantly reduces in vivo injectability failures. The injectable compositions can be made by mixing dry microparticles with an aqueous injection vehicle to form a suspension, and then mixing the suspension with a viscosity enhancing agent to increase the viscosity of the fluid phase of the suspension to the desired level for improved injectability.

    Abstract: The invention relates to a sustained release composition and methods of forming and using said composition for the sustained release of biologically active agent. The sustained release compositions of the invention comprise a biocompatible polymer and a biologically active agent characterized by a porous center and a less porous outer layer wherein the center and outer layer consist of essentially the same materials. The sustained release compositions can be prepared by annealing at least a substantial portion of the exterior surface of a polymer/active agent matrix. The compositions which have been annealed exhibit a decrease in the release of agent over the first 24 hours following administration (i.e., reduced burst) and as a result can show an increase in the duration of sustained release thereby providing increased therapeutic benefits.

    Abstract: Method and apparatus for preparing microparticles using liquid-liquid extraction. A first phase and a second phase are combined to form an emulsion. A portion of the second phase is separated from the emulsion (solvent rich), and the solvent is extracted from the separated second phase, which is then returned (solvent poor) to the emulsion. This process of separation of a solvent rich phase, extraction of solvent, and return of a solvent poor phase, is carried out until a selected level of solvent in the emulsion is achieved. Alternatively, the separated solvent rich phase is not returned to the emulsion, but replaced with another solution, such as an aqueous solution, that is free from solvent. The solvent is preferably extracted into an extraction liquid that functions as a “solvent sink” for the solvent.

The continuous phase can comprise one or more polymer solvents, a polymer solvent/polymer non-solvent mixture, or a polymer solvent/active agent non-solvent mixture. When the continuous phase comprises a polymer solvent/active agent non-solvent, the active agent can also be present as a microparticulate rather than in solution. The continuous phase is then removed from the polymer/active agent solution, thereby forming a solid polymer/active agent matrix.

    Abstract: A method of purifying human growth hormone (hGH) from deamidated hGH, oxidized hGH or both. The method comprises the steps of forming a metal cation complexed-hGH composition enriched in native hGH under conditions wherein none or less than all of the deamidated hGH, oxidized hGH or both complex with a metal cation and, isolating the metal cation-complexed hGH composition from the deamidated hGH, oxidized hGH or combination thereof. The method can further comprise the step of releasing purified hGH from the complex. Alternatively, the metal cation-complexed hGH can be encapsulated into a biocompatible polymer for sustained release of hGH. The purification method can be performed one or more times using the hGH released from the metal cation-complexed hGH composition, depending on the purity of the hGH desired.

The method for preparing micron particles of a labile agent comprises forming a suspension comprising the labile agent, dispersed in a polymer solution comprising at least one biocompatible polymer and at least one polymer solvent, and wet milling of the suspension.
