Abstract: A large scale cell harvesting method for a pack-bed culture device by providing cell growing space that is packed with a plurality of porous matrices and/or matrix means. When a cell detaching step at least comprising shaking or spinning the culture chamber is performed, the unoccupied space is large enough for the porous matrices collide to each other or to the culture chamber to generate momentum whereby the cells detach from the porous matrices to harvest the cells.
