Abstract: The present disclosure describes methods of preparing stable, solubilized silk fibroin preparations. The methods include providing a processed silk fibroin preparation. preparing a solution of the processed silk fibroin preparation in an aqueous TRIS buffer to provide a fibroin solution, treating the fibroin solution at least at a first pressure of 0 to 30 psi and a first temperature of 100° C. to 135° C. for a first time of 1 to 90 min to provide the stable. solubilized silk fibroin preparation. The stable, solubilized silk fibroin preparation has a molecular weight that changes by less than 33% during the first and optional second treating step, does not form a gel during the first and optional second treating step, remains clear after the first and optional second treating step, or a combination thereof.
Abstract: Described herein is a method of purifying a silk fibroin preparation including preparing an aqueous silk fibroin solution having a concentration of greater than or equal to 5% w/v silk fibroin from the silk fibroin preparation, wherein the silk fibroin preparation includes a chaotropic salt; and exchanging the chaotropic salt from the silk fibroin solution at a pH below the isoelectric point of the silk fibroin, wherein the pH is between 2 and 5, replacing the chaotropic salt with a buffer comprising 10 to 300 mM of a second salt, or a combination thereof, to prepare the purified silk fibroin.
Abstract: The embodiments disclosed and embraced by the present invention include silk-based products useful in the treatment, diagnosis, palliation, and/or amelioration of ocular diseases or conditions of the eye including those of the structures of the eye and surrounding tissue. Such silk-based products may effect beneficial outcomes alone or in combination with therapeutic modalities, compounds or medicaments.
