Abstract: Devices, compositions, and methods for handling, separating, packaging, and utilization of spermatozoa (1) that can be derived from previously frozen sperm samples collected from a male mammal. Specifically, techniques to uniformly stain (2) spermatozoal DNA even when derived from previously frozen sperm and separation techniques to separate and isolate spermatozoa even when derived from previously frozen sperm samples into X-chromosome bearing and Y-chromosome bearing populations having high purity.

Abstract: A novel ACC synthase gene in geranium PHSacc-25. PHSacc-25 was identified by amplifying isolated mRNA using polymerase chain reaction, the resulting cDNAs were cloned into a plasmid, positive clones were isolated, and the cDNA insert was partially sequenced. A portion of the DNA sequence encoding for the enzyme ACC synthase in geranium plants (specifically from Pelargonium hortorum cv Sincerity) was identified. The cDNA sequence corresponds to a gene which may be important in the control of ethylene production.

Abstract: Genes which encode ACC synthase are identified for the geranium plant, specifically Pelargonium x hortorum cv sincerity. These genes are shown as modified to achieve a transgenic plant which resists wilting and the like as a result of reduced ethylene production. This alteration is reproduced by the transformed plant. Isolation of high quality mRNA is achieved through use and adaptation of a 2-butoxyethanol precipitation technique using large amount of initial tissue in order to achieve critical mass for precipitation.
