  • Patent number: 5888544
Abstract: The effervescent system for effervescent tablets and/or effervescent granules contains, on the one hand, particles of a solid, edible, organic acid and, on the other hand, particles of at least one alkali metal bicarbonate, of which at least 1, preferably from 2 to 4, but at most 10, preferably at most 8, % by weight are superficially converted into dry alkali metal carbonate free of water of crystallization. The acid particles, preferably citric acid particles, can be covered in a manner known per se by partial reaction with at least one carbonate and/or bicarbonate, preferably from about 20 to 40% by weight thereof. The conversion mentioned is carried out by a procedure in which commercial alkali metal bicarbonate particles are heated in a vacuum vessel at less than 40, preferably less than 20, in particular less than 10, mbar at above 60.degree. C., preferably between 80.degree. C. and 120.degree. C., in particular at about 100.degree. C., and are cooled to below 60.degree. C., preferably to below 50.

  • Patent number: 5834019
Abstract: A pharmaceutical formulation includes a hydrophobic active substance, an effervescent system, and at least two surfactants. Each of the surfactants is selected from a different group selected from the following three groups: phospholipids, polysorbates, and esterified sugars. The formulations provide improved dispersion and bioavailability of hydrophobic active substances.

  • Patent number: 5831123
Abstract: A process for producing a granulated material from a powder mixture containing at least two components, one of which is in crystal or crystalline form and at least one contains water of crystallization and/or is reactive, granulation is carried out in an inclined drum. Powder is fed in at one end and granulate removed at the other, after being transported inside the drum for a desired period while the drum is rocked in a way which largely eliminates shear forces. A quantity of liquid at least approximately equal to the quantity of water of crystallization present and sufficient for the formation of a superficially active solution adhesive is released inside the drum, where appropriate by partial reaction of the reactive component(s), after which the powder mixture components are granulated by surface adhesion.

  • Patent number: 5792473
Abstract: In accordance with this invention there is provided a granular product with an effervescent system which comprises acid-sensitive pharmaceutically active substances, such as, for example, betacarotene, cimetidine, ranitidine or cisapride, which is specially useful to prevent antacid action, having an acid-neutralizing capacity below about 5 meq, at a weight of about 1.6 to about 2.3 grams. The effervescent grains are made from carrier crystals of at least one solid, edible organic acid, preferably citric acid and/or tartaric acid, and are present as a granular product, separate from the pharmaceutically active substance, and are coated with at least one layer of a neutral substance which is soluble in water and/or alcohol and which is able to bring about a melting point depression of the acid grains at their surface, such as, for example, a water-soluble polymer, a polyalcohol, a carbohydrate and/or a hydrocolloid.

  • Patent number: 5759575
Abstract: An effervescent granulated material for a pharmaceutical preparation contains calcium carbonate and citric acid. From 5-20 parts by weight of the total acid provided for the reaction with the calcium carbonate is replaced by at least one of malic acid, gluconic acid, lactic acid, and their salts. Citric acid and calcium carbonate are granulated with a partial reaction, and in each case at least part of the compound partially replacing the citric acid is granulated in solid, powdered form together with the citric acid and the calcium carbonate and/or dissolved in a granulation liquid or buffer solution.

  • Patent number: 5593693
Abstract: The effervescent mixture contains at least one acidic, insoluble or slightly soluble pharmaceutical active ingredient in the form of its soluble alkali metal salt or lysinate, which is present as an intimate mixture with the alkaline component and preferably coats the particles of the alkaline component of the effervescent system, for example sodium bicarbonate and/or sodium carbonate. Both components in very finely powdered form may also be present as granules, with the aid of a binder. In particular, the effervescent mixture contains, per 100 parts of active ingredient, at least 0.5 part, preferably 1 to 3 parts, of a surfactant, in particular dioctyl sulfosuccinate, and/or at least 1 part, preferably 3 to 5 parts, of a binder or suspending agent, in particular polyvinylpyrrolidone. The acidic component of the effervescent system is passivated on the surface.

  • Patent number: 5527540
Abstract: In the pharmaceutical formulation of at least one alkali-sensitive active substance with an effervescent system, the carbonate component is embedded in at least one edible, organic acid and is preferably covered by said acid or by another acid. The active substance is embedded in at least one of the following compounds: an edible, organic acid, a higher alcohol, a hydrocolloid or a relatively long-chain polyvinylpyrrolidone, preferably covered with at least one of the stated compounds. The contact zone between active substance and effervescent system should have a pH of not more than 4.5. Both effervescent system particles and active substance particles, which are embedded or optionally covered in this manner, may be applied to carrier crystals of the same or another acid. The mixture is preferably pressed to give tablets. The acid provided for the embedding or covering may contain 0.1 to 3 mg of ethylenediaminetetraacetic acid per tablet.

  • Patent number: 5415870
Abstract: The effervescent tablet contains an effervescent system composed of carrier crystals of at least one solid, edible, organic acid, at least one component which forms gas by reacting with the acid with salt formation and at least one salt formed from the acid and the gas-forming component. A first layer of a different acid and a second layer of a (preferably acidic) salt of at least one of the two acids are applied to the carrier crystals. Of the total amount of the acid(s) used and of the gas-forming components, 10 to 40, in particular 10 to 20, percent by weight are in the form of the salts, preferably in the form of acidic salts. Carrier crystals are, in particular, citric acid, malic acid, tartartic acid, monosodium citrate and/or ascorbic acid. The first layer may contain gluconic acid delta-lactone and is preferably covered with citric acid, malic acid and/or tartaric acid.

  • Patent number: 4968517
Abstract: The present invention relates to the formation of an effervescent granulate which contains carnitine. The granulate is formed by combining an edible organic acid with an alkaline carbonate or an alkaline earth metal carbonate to form a preliminary reaction solution. After the addition of salt, the crystals are stirred and dried and then finally combined with carnitine.
