  • Patent number: 4415665
Abstract: The invention relates to a method of coupling organic ligands to a polymeric carrier. The organic ligand contains at least one primary or secondary amino group, at least one thiol group and/or at least one aromatic hydroxy group, and the polymeric carrier contains at least one hydroxy group. According to the invention the coupling is performed by forming a reactive derivative of the polymeric carrier by treatment with a sulfonyl halogenid, and reacting the formed reactive derivative with the organic ligand, which is thereby bonded directly to a carbon atom in the polymeric carrier. The polymeric carrier can, for example, be a possibly cross-linked polysaccharide, and the organic ligand is preferably a biological material such as a protein. The coupling product can, for example, be used for immunologic determinations, for affinity chromatography, etc. The coupling method can be performed under mild conditions not damaging sensitive organic ligands.

  • Patent number: 4389316
Abstract: A method and a device for applying a sample solution to a liquid chromatographic column. The device comprises a container of uniform cross section and a piston slidably arranged in said container and sealingly contacting the container wall so as to divide the container into a driving solution chamber and a sample solution chamber. The piston is arranged to be displaced towards the sample solution chamber when driving solution is pumped into the driving solution chamber while expelling sample solution from the sample solution chamber in an amount proportional to the amount of driving solution pumped into the driving solution chamber. When all sample solution has been expelled from the sample solution chamber the driving solution chamber is automatically put into communication with the outlet of the container.

  • Patent number: 4312739
Abstract: A medium for isoelectric focusing is produced which consists of or contains agarose into which positively charged substituents have been introduced to neutralize negatively charged groups inherently present in the agarose, said substituents containing as the only charged group a quaternary amino group with a positive charge independent of pH at least in the range 2-12. The medium may consist of a mixture of two or more agarose preparations with different degrees of substitution with regard to the introduced substituents. Unmodified agarose may also constitute one of the components of the medium.

  • Patent number: 4086222
Abstract: This invention relates to a new method of isolating highly purified albumin in high yields from plasma products.According to the invention a particular plasma fraction, containing albumin in dissolved form and being essentially free from the coagulation factors I, II, VII, VIII, IX and X and from the main part of IgG, is subjected to a two-step chromatographic separation using aqueous buffer systems. The first step is carried out on an anion exchanger and the second step on a cation exchanger, or vice versa. In a preferred embodiment a buffer of pH 4.5 - 4.9 and an ionic strength of 0.025 - 0.1 is used for the separation on the anion exchanger, whereas a buffer of pH 5.2 - 6.5 and an ionic strength of 0.1 - 0.05 is used for the separation on the cation exchanger.
