Methods of interfering with neuronal transmission comprising topical application of an effective amount of the at least one tricyclic 3,4-propinoperhydropurine are provided for treatment of a variety of indications that relate to muscle relaxation, anesthesia, muscle spasms and similar causes.

In another aspect of the invention, effective amounts of the pharmaceutical compositions and a transdermal therapeutic system are provided for transdermal administration of at least one tricyclic 3,4-propinoperhydropurine. The pharmaceutical composition contains at least one at least one tricyclic 3,4-propinoperhydropurine, and may be formulated for transdermal drug delivery. The transdermal drug delivery system may be a laminated composite comprising a backing layer, a drug reservoir, and a means for affixing the composite to the skin.
