Abstract: The colorimetric bio sensor includes: an upper housing including a blood cell separation module separating blood plasma from blood injected from the outside and spreads the separated blood plasma and a specimen delivery hole being located on the lower end of the blood cell separation module and formed to penetrate the upper housing, so that the lower end of the blood cell separation module is exposed to the outside; and a lower housing including a membrane containing a colorimetric reaction substance causing a colorimetric reaction with a target substance in the blood plasma by being in contact with the blood cell separation module through the specimen delivery hole, and a protrusion being located on the lower end of the membrane and protruding to a height corresponding to the depth of the specimen delivery hole.
