    Abstract: A measuring method for low concentration nucleic acid sample is provided. The measuring method is a nucleic acid measuring method for samples of low nucleic acid concentration range in a qPCR experiment, so as to extend the linear dynamic range of experimental detection and increase detecting sensitivity by correcting the original Cq value. The method includes providing a test plate with a plurality of reaction wells or holes, so as to perform qPCR reaction on the nucleic acid samples. Next, performing the adjustment step based on the positive well measurement value derived from the number of positive wells, so as to correct the Cq value of qPCR reaction to the expected value of linear range.

    Abstract: A method of multi-gene absolute quantification for PCR array is provided, wherein a nucleic acid sample to be tested includes at least one kind of nucleic acid target. Primers for amplifying a plurality of standard DNA or the nucleic acid target to be tested are respectively disposed in a plurality of reaction wells of a test carrier. Afterwards, the plurality of standard DNA with known copy number and the nucleic acid sample are mixed and added into the reaction wells, and qPCR is performed on the standard DNA and the nucleic acid sample. A sequence-specific combination of a forward primer and a reverse primer is designed for each standard DNA, and the same DNA template sequence is presented between regions corresponding to the forward primer and the reverse primer. The primers for amplifying standard DNA and the nucleic acid target to be tested have similar amplification efficiencies.

A qPCR master mix, a forward primer, a reverse primer, a hybridization probe, a double-stranded DNA binding dye, and nucleic acid samples are added into reaction wells for qPCR reaction, and the fluorescence intensity of each of a hybridization probe and a double-stranded DNA binding dye varying with cycle number is respectively measured. Afterwards, amplicon length is obtained by applying a calculating method.

    Abstract: A multiplex slide plate device and an operation method thereof are provided. The multiplex slide plate device includes a slide plate and a sacrificial layer. The slide plate has reaction vessels arranged in an array, an injection hole and an exhaust hole, wherein each of the reaction vessels has an opening portion and a bottom portion. The sacrificial layer has a microfluidic channel, wherein the microfluidic channel has an injection channel, a main channel and a distal channel connected to each other. The sacrificial layer is assembled to the slide plate, wherein the main channel faces the opening portion. A sample solution is injected into the injection channel, such that the sample solution flows from the injection channel through the main channel to the distal channel, wherein the sample solution loads into each of the reaction vessels while flowing through the main channel.
