Abstract: Effective polymeric transfection reagents for delivery of nucleic acids and their complexes to modify host cells, particularly hematopoietic cells including myeloid and lymphoid cells, pharmaceutical compositions comprising same, and methods of preparing and using same are provided. An effective compound comprises a low molecular weight polymer and aliphatic lipid-thioester or lipid-ester groups. A nanoparticle comprises the compound complexed with a nucleic acid and/or an additive. A composition or pharmaceutical composition comprises the nanoparticle and a pharmaceutically acceptable carrier. A method of treating, preventing, or ameliorating a disease in a subject comprises administering to the subject an effective amount of the nanoparticle or the composition or pharmaceutical composition.
