Publication number: 20070227891Abstract: Notable techniques for protein separation prior to any further downstream analysis include the Sodium Doecyl Sulphate-PolyAcrylamide Gel Electrophoresis (SDS-PAGE). However, SDS-PAGE suffers from limitations such as band broadening and the ineffective separation of proteins or proteins isoforms with very similar migration mobilities under the influence of an electric field. Currently, the best method for protein separation and resolution with very narrow molecular weight variation utililizing SDS-PAGE is by pulse electrophoresis. However, pulse electrophoresis introduces new limitations such as the long run period required, band broadening contributed by diffusion when the electrical field is switched off, and the need for casting of inconvenient and unconventional long separating gel arise.
