According to a method of subfractionation culture and proliferation of stem cells of the present disclosure, it is possible to obtain a large quantity of desired monoclonal stem cells in a short time by rapid proliferation of monoclonal stem cells, and the monoclonal stem cells obtained thereby are stem cells with enhanced therapeutic effects on graft-versus-host disease, and thus can be beneficially used as a graft-versus-host disease therapeutic agent.
