Publication number: 20130052718Abstract: The invention provides self replicating infectious recombinant paramyxoviruses. The recombinant paramyxovirus preferably have one or more attenuating mutations. In some embodiments, the recombinant paramyxovirus has a separate variant polynucleotide encoding a P protein and a separate monocistronic polynucleotide encoding a V protein. In some embodiments, recombinant paramyxovirus have at least one temperature sensitive mutation and one non-temperature sensitive mutation. Also provided are compositions and methods for using the recombinant paramyxoviruses as described herein.Type: ApplicationFiled: January 10, 2006Publication date: February 28, 2013Applicant: THE GOVERNMENT OF THE US, REPRESENTED BY THE SECRETARY , DEPT. OF HEALTH AND HUMAN SERVICESInventors: Mario H. Skiadopoulos, Peter L. Collins, Sheila Nolan

Publication number: 20110318737Abstract: Materials and processes are provided for the detection of Legionella species in a sample. The inventive process includes exposing a sample to a forward primer and a reverse primer to yield an amplicon. The amplicon is detectable by at least one probe. The inventive process detects multiple species of Legionella bacteria or is specific for Legionella pneumophila. A kit is provided with inventive primers and probes for the detection of a Legionella species in a sample.Type: ApplicationFiled: December 17, 2009Publication date: December 29, 2011Applicant: The Government of the US as Represented by the Secretary of the Dept.of Health and Human ServicesInventors: Robert F. Benson, Brian P. Holloway, Karen A. McCaustland, Patrick Genyan Yang

Publication number: 20100055670Abstract: The invention provides recombinant Hepatitis A Virus (HAV) nucleic acids and host cells that are permissive for their growth and replication. The recombinant Hepatitis A Virus nucleic acids not particularly limited, except that they incorporate at least one heterologous nucleic acid fragment. The heterologous nucleic acid can encode a selectable marker gene and such recombinant HAV nucleic acids are useful for selecting cells that are permissive for growth and replication of wild type HAV. Alternatively, the heterologous nucleic acid may encode a vaccine antigen or other expression product that is desirable to express in a cell harboring the recombinant HAV nucleic acid. The invention further provides cell lines permissive for growth and replication of wild type HAV or HAV having minimal mutations for growth in cell culture. The invention further provides methods for producing HAV vaccines and for monitoring environmental and patient samples for the presence of HAV.Type: ApplicationFiled: June 26, 2006Publication date: March 4, 2010Applicant: The Government of the US, Represented by the Secretary, Dept. of Health and Human ServicesInventors: Gerardo Kaplan, Krishnamurthy Konduru
