Abstract: Monoclonal antibodies are described which are potent reagents for the immunologic definition of hairy leukemic cells.S-HCL 1 is a pan-B cell marker with markedly increased staining on hairy cells versus normal B-lymphocytes. It also stains with a variety of B-cell leukemias and lymphomas, but is not crossreactive with any other cell type in man. With these features, it is also a useful reagent for the detection of human B-lymphocytes.S-HCL-3 monoclonal antibody recognizes an antigen present on entirely different cell lineages, namely macrophages of almost all tissues and polymorphonuclear cells. This antigen is not expressed on any other malignant lymphocytes except hairy cells. Moreover, this antigen is not found on other non-Hodgkin lymphomas which may resemble hairy cell leukemia. Accordingly, b-HCl-3 is useful in the diagnosis of hairy cell leukemic.

Abstract: A method for determining the presence of a predetermined minimum detectible amount of one or more analytes in a sample suspected of containing the analyte is disclosed. Each analyte is a member of a specific binding pair ("sbp member") consisting of ligand and its complementary receptor. The method comprises contacting with a test solution containing the sample and predetermined amounts of two or more of a plurality of first sbp members, each respectively analogous to one of the analytes, a contact portion of a piece of bibulous material capable of being traversed in at least one direction by the test solution by capillary migration. The bibulous material contains predetermined amounts of two or more of a plurality of second sbp members, each respectively capable of binding one of the analytes and corresponding first sbp member.

Abstract: In accordance with the invention there is provided a Petri dish comprising a container for the growth of microorganism cultures and a lid for the container. The construction of the lid and the container provides an improved stacking feature when a plurality of Petri dishes are stacked. Abstract: Sulfate reducing bacteria are detected by preparing a lysate so as to release an enzyme essential to derive energy by reduction of sulfate such as adenosine 5'-phosphosulfate reductase (APS reductase), containing the lysate with an antibody for the enzyme, and detecting the presence of the reaction product of the enzyme and the antibody. In one aspect, sulfate reducing bacteria are controlled responsive to such assay. In another aspect, a "test kit" is provided for carrying out the assay.

Abstract: Use of labeled monoclonal antibody 50 H.19 to detect the location and concentration of blood platelets in a host is disclosed. MAb 50 H.19 labeled with a label which can be detected from without the host (e.g. a radiolabel or NMR-label) is intravenously injected to immunoreact with platelets in vivo or is first immunoreacted in vitro with platelets, which are then intravenously injected into the host. By means of the labeled MAb 50 H.19 bound platelets it is possible to detect and locate thrombi, emboli, atherosclerotic obstructions, bacterial endocarditis and to do spleen imaging. In another embodiment of the invention, the MAb 50 H.19 is bound to an enzymatic thrombolytic agent and injected intravenously to permit the therapeutic application of the agent directly to thrombi.

Abstract: Microbial activity in an aqueous medium is measured in a bioelectrochemical cell in which the working electrode comprises an electrically conductive carbon-bearing porous material; suitably a graphitized carbon felt or cloth. This enhances the BEC response by increasing the surface area and by absorbing microbes onto the surface; and the electrode material can also be used to preconcentrate the microbes prior to use as an electrode in the cell.

Abstract: A novel acetylpolyamine amidohydrolase is described. The enzyme specifically hydrolyzes the amino bond in acetylputrescine, acetylcadaverine, acetylspermidine and acetylspermine with strong substrate affinity. The enzyme is preferably produced by culturing a microorganism belonging to the genus Mycoplana, and is used in the quantitative determination of polyamine contained in a living body sample, which is useful for cancer diagnosis.
Abstract: Novel Fc receptors, denoted type VI, are disclosed as reacting with rat immunoglobulins with a reasonable affinity. The type VI receptors are isolated from Streptococcus zooepidemicus strain s212 (ATCC 53698) and Streptococcus zooepidemicus strain RSS-212 (ATCC 53697) culture supernatants. These receptors, or the microorganisms which produce them, are useful in immunoassays.

Abstract: Methods and materials for preparing specific binding reagents with a multiplicity of relatively noninterfering label moieties are described. By spacing the labels at the surface of a specific reagent with bulking agent, increased sensitivity can be achieved without interference between individual labeling entities.
Abstract: The present invention involves a method for producing a substrate useful in a system for the detection of antibodies directed against platelet antigens. This method comprises several steps. A platelet sample of interest is initially treated with an aqueous solution comprising a dialyzable nonionic detergent. This initial treatment is under conditions to solubilize platelet components and produce a platelet lysate. Such conditions may involve treatment of a platelet sample with an aqueous solution comprising nonionic detergent at a concentration between about 0.2% and about 0.5%. Platelet antigens are most preferably solubilized for about 30 min and at about 0.degree. C. in an aqueous solution comprising about 1 mg dialyzable nonionic detergent per mg platelet protein.The partially purified platelet antigens resulting from these manipulations are then preferably affixed to a solid matrix.

Abstract: Hybrid cell lines (hybridomas) which produce and secrete monoclonal antibodies with three distinct patterns of recognition are described. The first specificity pattern is defined by antibodies which (1) recognize Kunitz trypsin inhibitor (KTI), one of the principal protease inhibitors found in soybeans, but do not detect the Bowman-Birk inhibitors (BBI), the other major class of protease inhibitors in soybeans; (2) bind to native KTI isoforms a and b but do not react with KTI isoforms a and b which have been denatured by moist heat or alkaline treatment or which have been subjected to disulfide exchange; and (3) do not recognize native KTI isoform c.

Abstract: The sample for analysis containing the co-enzyme to be detemined, inter alia NADH or NADPH, is placed in the presence of a fluorinated aromatic compound. In the presence of oxygen and peroxidase, the fluorinated compound releases fluoride ions at a rate proportional to the quantity of co-enzyme to be determined. Abstract: A method is disclosed for the immunolocalization of epitopes of non-glucidic nature, in cellular cultures, in tissue slices, or immobilized on such supports as nitrocellulose, by means of the incubation of the sample with specific antibodies for the antigen to be immunolocalized, belonging to the class of the IgM's, and sequential treatment of concanavalin A and with an enzymatic marker capable of binding with concanavalin A, such as, e.g., peroxidase. The method comprises, before the incubation, a suitable treatment of the sample under test, e.g., with periodic acid, in order to prevent concanavalin A from binding with the carbohydrates present in the sample.
Abstract: Gas-generating activity of microorganisms is detected by providing a vessel in which the microorganisms are incubated, the vessel having a cap with a membrane indicator therein. The membrane has a deformed region which is inflated to indicate the existence of an increase in pressure within the vessel as compared with ambient pressure. Visual or automatic monitoring of the inflated portion is disclosed.
Abstract: An assay to detect the presence of live non-A, non-B hepatitis virus in vitro. Bone marrow cells are exposed to a body fluid or biological preparation to be tested and the cells are placed in suspension. When using bone marrow cells, growth factors are added to the bone marrow stem cells. Therefore, if the number of colonies growing in the mixture are less than that number present in the culture of cells exposed to a sample that has been determined to contain no live virus, live non-A, non-B hepatitis virus is present in the sample tested. The assay is particularly useful to determine the presence of live non-A, non-B, hepatitis virus in a vaccine.

Abstract: Serum samples for vitamin B.sub.12 analysis can be pretreated using a combination of peroxy acid and dithiothreitol. The pretreatment makes vitamin B.sub.12 bound to serum binding proteins available for analysis by any of a variety of currently available assay techniques. The pretreatment finds particular use in an assay using solid phase intrinsic factor as a specific binding protein.
