The invention further relates to novel processes of preparing novel stem cell lines which are free of animal products, feeder cells, growth factors, leukaemia inhibitory factor, supplementary mineral combinations, amino acid supplements, vitamin supplements, fibroblast growth factor, membrane associated steel factor, soluble steel factor and conditioned media. This invention further relates to the isolation, culture, maintenance, expansion, differentiation, storage, and preservation of such stem cells.

    Abstract: The present invention provides a method for purifying bacterial minicells that involves subjecting a sample containing minicells to density gradient centrifugation in a biologically compatible medium. The method optionally includes a preliminary differential centrifugation step and one or more filtration steps. The invention also provides a method for purifying bacterial minicells in which a sample containing minicells is subjected to a condition that induces parent bacterial cells to adopt a filamentous form, followed by filtration of the sample to separate minicells from parent bacterial cells. The inventive methods optionally include one or more steps to remove endotoxin from purified minicell preparations, and/or treatment of purified minicell preparations with an antibiotic. Additionally, the invention provides purified minicell preparations, prepared according to the foregoing methods, and containing fewer than about 1 contaminating parent bacterial cell per 107, 108, 109, 1010, or 1011 minicells.

    Abstract: The present invention relates to a layered cell sorted tissue that is formed in vitro. The tissue is generated by the spontaneous sorting of cells from a homogenous cell mixture into discrete layers by cell type. Connective tissue components, such as fibronectin, may be used to manipulate orientation of the layers during the cell sorting process. The layered cell sorted tissue may be used as a skin graft for burns, wounds, and ulcers. The tissue may also be used in assays to determine effects of chemicals or drugs on human tissue in vitro as well as provide an in vitro assay for tumor cell metastasis.
