  • Publication number: 20110027911
Abstract: Provided are methods for solubilizing a membrane protein in order to provide substantially homogeneous membrane proteins that are solubilized within reverse micelle systems at concentrations that are suitable for analytical study of such membrane proteins, including nuclear magnetic resonance spectroscopy. Also provided are substantially homogeneous membrane proteins that are solubilized within a reverse micelle system, as well as methods for the study of solubilized membrane proteins, and for the screening of drug candidates that target such membrane proteins.
    Type: Application
    Filed: January 9, 2009
    Publication date: February 3, 2011
Applicant: THE TRUSTEES OF THE UNIVERSITY OF PENNSYLVANIA
Inventors: A. Joshua Wand, Ronald W. Peterson, Joseph M. Kielec
  • Patent number: 6486672
Abstract: The present invention provides a method for reducing the correlation time for global tumbling for analysis of proteins and other macromolecules by NMR spectroscopy. The method comprises placing a macromolecule, hydrated and without significant structure perturbation, within a reverse micelle in a suitable fluid medium of low viscosity. A high pressure NMR cell for use with the method of the present invention is also disclosed.
    Type: Grant
    Filed: January 25, 2001
Date of Patent: November 26, 2002
Inventors: A. Joshua Wand, Mark R. Ehrhardt, Peter F. Flynn
