Patent number: 10857483Abstract: Disclosed is a continuous process for separating or extracting proteins from a low grade mixture of a protein of interest, other proteins, impurities, and salts in a continuous simulated moving bed separation process. The invention provides for direct extraction of heme protein and plant protein from a crude mixture of such proteins, other proteins, impurities and salts using the chromatographic technique of simulated moving bed (SMB) continuous chromatography. The SMB process combines the steps of feed loading, adsorbent washing, product elution, adsorbent regeneration, and adsorbent equilibration. The novel strong anion exchange resin adsorbent is a quaternary amine cross-linked microcellulose wherein the microcellulose is cross-linked with epichlorohydrin and the quaternary amine is 2,3-epoxypropyltrimethyl-ammonium chloride which exhibits selective adsorption of proteins and complete regeneration.Type: GrantFiled: November 20, 2017Date of Patent: December 8, 2020Assignee: Orochem Technologies Inc.Inventors: Anil R Oroskar, Babu Siddegowda Antharavally, Anantha Krishna Mallia

Patent number: 9452968Abstract: Disclosed is a process for the separation and purification of a diacid from a fermentation broth withdrawn directly from a fermentation zone for producing the diacid, wherein the fermentation broth comprises the diacid and water, a corresponding monoacid, and a corresponding ?-hydroxyacid. The process comprises passing the fermentation broth to a filtration zone having a filtration media and filtering the fermentation broth to provide a filtered feedstock. The filtered feedstock and a desorbent stream are passed to a simulated moving bed separation zone (SMB) operating in reverse phase at effective SMB conditions. The diacid may be adipic, acid, a 6-carbon atom diacid, or dodecanedioic acid, a 12-carbon atom diacid. The process is useful in simplifying the manufacturing the manufacture of diacids produced by fermentation. Adipic acid is used as monomer for the production of nylon.Type: GrantFiled: April 22, 2015Date of Patent: September 27, 2016Assignee: OROCHEM TECHNOLOGIES, Inc.Inventors: Anil R. Publication number: 20030111345Abstract: A method is disclosed for the detection of a His-tagged target biomolecule while said molecule remains in the gel in which it has been separated from other constituents by electrophoresis. The method involves, while the separated His-tagged biomolecule remains in the gel, forming a chelate between the His-tagged molecule, a lanthanide metal ion which exhibits fluorescence, and a sensitizer which enhances the fluorescence of the ion. The gel is then exposed to a u.v. light source to thereby generate a fluorogenic signal indicating the presence of the chelate and, in turn, the His-tagged biomolecule. The fluorogenic signal can then be detected either by direct visualization and/or by other means, such as photographically, e.g., film or charged coupled device (CCD) imaging.Type: ApplicationFiled: January 28, 2003Publication date: June 19, 2003Inventor: A. Krishna Mallia

Patent number: 5538858Abstract: A rapid radioactive method of measuring enzymatic activity of a protein kinase is disclosed. The method is an improvement to existing methodology which involves phosphorylating a peptide substrate using .sup.32 P-ATP, adsorbing the phosphorylated peptide to a solid phase, washing the phase to remove non-adsorbed .sup.32 P-ATP, and measuring the radioactivity of the phosphorylated peptide adsorbed to the phase. The disclosed improvement uses a membrane as the solid phase and positions the membrane within a chamber to separate the chamber into a first and second region. Washing is accomplished with centrifugal force; the washed solution being forced through the membrane from the first region into the second region.Type: GrantFiled: April 8, 1994Date of Patent: July 23, 1996Assignee: Pierce Chemical CompanyInventors: A. The method is an improvement to existing methodology which involves phosphorylating a peptide substrate, adsorbing the phosphorylated peptide to a solid phase, washing the phase to remove non-adsorbed constituents, and measuring the amount of phosphorylated peptide adsorbed to the phase. The disclosed improvement uses a membrane as the solid phase and positions the membrane within a chamber to separate the chamber into a first and a second region. Washing is accomplished with centrifugal force; the washed solution being forced through the membrane from the first region into the second region.
